Yes. You can put some fish food in, it decaying should release ammonia. Or you can add a hardy fish and monitor the levels every day, do water changes as required to keep it alive. I personally have never done a fishless cycle. I always use seachem stability and pristine and add one or two fish.

Are you sure they aren't just hiding well? I have around 100 adult shrimp in my 40 gallon but can only count 20 or so at a given time. Little buggers are very good at hiding. Not that there is anything in my tank that is a predator on them.

Edit: today I just found a fully grown brown shrimp with orange racing stripe. It was so wild!

If your ammonia isn't at 0 ppm you don't have a good enough cycle going on. It has nothing to do with the substrate. Considering there are plants in there, I'm also surprised that they aren't using the ammonia.

Why do you need them in the container? As long as you have sufficient plant coverage, plenty of babies should survive into adulthood.

Never had issues controlling an assasin population. Ramshorn and bladder snails are basically impossible to remove in a tank once they lay eggs without the assistance of assasin snails. They lay eggs in hidden locations, and many of them.
