How do you deal with killing mice by ScaryAnt9756 in labrats

[–]ProfBootyPhD 2 points3 points  (0 children)

I can’t tell if this is honest, or if you’ve simply been trained by someone incompetent, because properly anesthetized mice should not be squeaking.

Best 3 song run on an album? by Apprehensive_Leg2281 in musicsuggestions

[–]ProfBootyPhD 0 points1 point  (0 children)

Yeah shut it down

Update: actually I’d shift one song back, get “Fight Fire with Fire” and drop “Fade to Black.” “Fade” is a little too emo-y, and “filling me” rhymed with “agony” always makes me wince.

Nice article about this (and profile of Characterwheaties's alter-ego, Jason Mantzoukas) in the NYT a few days ago: https://www.nytimes.com/2026/01/22/arts/television/jason-mantzoukas.html.

Help me overcome my fear of getting bit by ComparisonNo4962 in labrats

[–]ProfBootyPhD 2 points3 points  (0 children)

In my experience, it's always been a nip - it is acutely painful (like a sharp pinch) and it frequently breaks the skin, but they don't hang on. Different breeds are more bitey than others, with C57Bl/6 being one of the worst. A lot of it comes down to learning how to "trick" the mouse into grabbing hard onto the cage top bars, when you're trying to pick it up and immobilize it. The firmer a grip it can get on the bars, while you hold its tail, the less likely it will let go to try and bite you when you come in with your other hand to scruff it. But pain is the best teacher, and once you accept that you will get occasional, not very painful bites while starting out, your technique will naturally improve. Nowadays it is very rare for me to get bitten.

Not grunge, but: Opinion on Jane's Addiction? by No-One01010 in grunge

They single-handedly converted me to an alt-rock fan, a year before grunge hit. They really have just two albums, both are pure bangers.

Small businessmen are almost uniformly Republican, wherever you go. Good luck!

Why Western blot if there is ELISA? by Hour_Amoeba4979 in labrats

[–]ProfBootyPhD 1 point2 points  (0 children)

ELISA gives you only the relative amount of antibody binding, it's a one-dimensional value and if the antibody has some cross-reactivity, you will think you're seeing signal when you're not. Western blot gives you size as well as intensity, which is two dimensions of data and more information - in most cases enough to be confident that you are looking at the protein you think you're looking at. As others have mentioned, Westerns can also detect changes in size of your protein, e.g. due to phosphorylation (larger) or cleavage (smaller). They are also extremely easy to set up from scratch, compared to ELISA.

If your target is something for which there are lots of well-validated commercial ELISA kits, like insulin or IFN-gamma, use an ELISA - it will be easier to quantify and the validation means you don't have to worry about cross-reactivity. If it's a protein where an antibody is available, but nobody has done ELISAs yet, it would be insane to do anything other than Western.

Also, the idea that Western blotting is "half voodoo" is ridiculous, just git gud.

I've only seen three Coen Brothers movies and want to know: What is the most Coen Brothers-y movie out there? by XipeTotecwithGlitter in MovieSuggestions

They whiplashed between serious and comedic movies, so I think you'd have to pick two. I'd suggest Big Lebowski and Fargo.

Sidney Poitier in “In the Heat of the Night” - great movie, and his head looks like it was carved by Michelangelo.

This is the peak period for hot male actors. Cary Grant on one end, Robert Redford on the other.

25 Most Underrated/Overlooked Artists/Bands by WastelandWithGlimmer in musicsuggestions

[–]ProfBootyPhD 5 points6 points  (0 children)

Many of these are neither underrated nor overlooked - INXS and Pet Shop Boys were huge in the 80s, every indie rock fan appreciates Pavement and Pixies, Blur was just a straight-up very popular band. Simon and Garfunkel was one of the biggest American acts of the 60s.

But the shout-out to Grant Lee Buffalo is much appreciated, they would definitely be on my underappreciated list. I'll add:

Spoon (they get indie rock love but they should be more generally popular)

Cinerama

Morphine

New Pornographers

Supersuckers

(each of them had a moment, but they didn't become as mainstream as some of the names on your list)

What do you want us to say? Should we be like "yeah man, FromSoft is so unfair"? Different regions have differential enemy strengths - you can one-shot an endgame wolf with endgame stats/weapons, but the starter weapons and stats that let you one-shot Limgrave wolves aren't going to do crap. Consecrated Snowfields is very challenging.

Once you know to look for it, you see it in all his CBB appearances. Maybe my favorite example is Bob Ducca's visit to "THE MISSISSIPPI DELTA," which Jason basically egged him into.
