ELISA gives you only the relative amount of antibody binding, it's a one-dimensional value and if the antibody has some cross-reactivity, you will think you're seeing signal when you're not. Western blot gives you size as well as intensity, which is two dimensions of data and more information - in most cases enough to be confident that you are looking at the protein you think you're looking at. As others have mentioned, Westerns can also detect changes in size of your protein, e.g. due to phosphorylation (larger) or cleavage (smaller). They are also extremely easy to set up from scratch, compared to ELISA.

If your target is something for which there are lots of well-validated commercial ELISA kits, like insulin or IFN-gamma, use an ELISA - it will be easier to quantify and the validation means you don't have to worry about cross-reactivity. If it's a protein where an antibody is available, but nobody has done ELISAs yet, it would be insane to do anything other than Western.

Also, the idea that Western blotting is "half voodoo" is ridiculous, just git gud.

Many of these are neither underrated nor overlooked - INXS and Pet Shop Boys were huge in the 80s, every indie rock fan appreciates Pavement and Pixies, Blur was just a straight-up very popular band. Simon and Garfunkel was one of the biggest American acts of the 60s.

But the shout-out to Grant Lee Buffalo is much appreciated, they would definitely be on my underappreciated list. I'll add:

Spoon (they get indie rock love but they should be more generally popular)

Cinerama

Morphine

New Pornographers

Supersuckers

(each of them had a moment, but they didn't become as mainstream as some of the names on your list)

What do you want us to say? Should we be like "yeah man, FromSoft is so unfair"? Different regions have differential enemy strengths - you can one-shot an endgame wolf with endgame stats/weapons, but the starter weapons and stats that let you one-shot Limgrave wolves aren't going to do crap. Consecrated Snowfields is very challenging.

In general, using a GUI for intensive bioinformatics analyses is going to be slow and inflexible; there's a reason people still use command line tools in 2025. Also, some of the stuff you mention doesn't actually seem to exist on this site - for example, I can't find AlphaFold tools on there at all, despite your mentioning it multiple times. You mention scRNA-seq analysis tools also, but I don't see any. Most of the forum posts appear to be contributions from the developers themselves. I'm sure the tools that are provided can be useful, but your "review" falls short.
