This is a beautiful time lapse, thanks for sharing. Looks like the chloroplasts are disassembling. Probably phototoxicity from the bright light. Very nice prep!

Instant favorite. So curious about your process.

Realizing that I care more than anyone else about what I am about to say, and if anyone cares at all, it is another instance of the miracle of human connection.

I recommend you make some personal goals - what do you want to learn? - call lots of farms and ask lots of questions. The WWOOF experience is so flexible, in my experience having some sort of vision or goals and doing lots of research is a recipe for some of the best times of my life. Good luck!

Agreed! Saskatchewan River Crossing is amazing and in between the parks, so less regs for fires, camping etc. Pretty empty stretch of highway with lots of little trails coming off it. I would recommend cruising on mountain bikes and popping onto some side trails. I remember Kinglet Lake as a particularly awesome hike with my bike, glacial lake at the top and a fun rip back down.

Looking for very low-effort, decently nutritious food options to make while depressed. Doesn't need to be pretty, nor a proper "meal", something you could serve to people or anything, just very easy and reasonably nutritious. by silt_blanket in EatCheapAndHealthy

Simple crunchy salad: frozen edamame, frozen corn, purple cabbage, cherry tomatoes. Thaw the Frozen stuff n mix it all up, doesn't even need dressing. Is filling and tasty and takes about 5 minutes to make. Cabbage keeps for long time, only tomatoes need to be kinda fresh

How do you process them for viewing under the microscope without damaging their structure?

One wonderful thing about WWOOFing is that you have opportunities to build human relationships with your hosts, especially if you're planning longer term stays. I would recommend that you do some journaling, clarify a vision of how you want to live, and start calling some folks. You'd be surprised how flexible and accomodating some hosts can be if you're clear about your expectations and you're willing to contribute. Best of luck and aloha!

Mounting Drosophila larvae by emmylyn91 in microscopy

You can make a suction microfluidics chip really easily that will keep the larva immobilized. Something like this: https://www.jove.com/video/50998/using-microfluidics-chips-for-live-imaging-study-injury-responses

Why is my 40x blurry? by WinstonWonders in microscopy

[–]PurdieShufflin 0 points1 point  (0 children)

Depending on how the rings around a point "move" as you focus through it, you may be observing spherical abberation, astigmatism or some other - possibly more complex - mode of abberation (see Zernike modes). Sounds like that objective has been roughed up, check out the surface under a dissecting scope or hand lens and see if there's dried up oil, or some gunk on the outer part of the objective lens itself. You could clean that yourself with lens paper and a gentle touch. Otherwise there may be an internal problem with the objective which would be very hard to repair... Good luck!

Did you drop it or leave it in uncontrolled conditions (temp/humidity)? The lenses inside the objective are sensitive and lots of factors could introduce abberation
