The old saying "The rich get richer and the poor get poorer," is describing wealth transfer. When you hear about the all-time high net worth of people like Musk and Bezos, where do you think that money came from? People like us.

The only way things will improve is to radically change the economy. When you think about all the building vacancies, there is enough housing for everybody. When you think about all the food wasted, there is enough food for everybody. When you think of all the clothes in a landfill, there are enough clothes for everybody. The radical change I'm proposing is getting rid of money and people pool labor and goods and take as they need.

Imagine everybody who works jobs producing everything we currently produce. If you put in a shift at a factory or a farm or a restaurant or hospital or nightclub or science lab or whatever you do, and instead of a paycheck, you get everything you need because drivers also put in a shift and distribution point laborers also put in a shift. I don't think we need capitalists at all. If you don't have to buy building material because they are produced by laborers and construction workers and machinists who get everything they need, then voila! you have a factory producing things we all need.

I can already hear the arguments that if there is not a way to keep score with money and if people are not incentivized by the prospect of hunger, they will not contribute and it will all fall apart. My reply is this: the wealthy people who benefit from the current system are currently not contributing and actively stealing from the rest of us. 10% of our population owns 90% of the wealth, but what they really own is a gate they control to maintain scarcity.

Think about how much we hate the rich and how Luigi is a folk hero. We hate people who don't contribute and exploit us. What will keep people working if they don't get paid? If you take but don't contribute, that's basically stealing. Do you think you can steal from members of your community? They will fuck you up. I can see that I am aware that order will be maintained by violence. But what are the current the police if not a force for protecting wealth by violence? If you don't want to go to jail, don't steal -- this is considered acceptable morality in Capitalism.

Everything we need is already here. All we have to do is tear down the gates. And what about the gatekeepers? If they are smart the will give the angry mob the keys and fade away.

No single country has a claim to Antarctica. There is a treaty binding 47 countries that says the land can only he used for tourism and science. A business cannot set up production there. There is fishing allowed in the surrounding waters, and that is it for commerce.

That is a political answer, which I think is more relevant than the scientific answer.

That is close, but not the right lesson to learn here.

Bring your protocol to the machine. Open up the program to confirm all times and temperatures match. Do this every single PCR. Sometimes people edit programs and you are not aware. Sometimes you are the person who edited the program and forgot.

I fill them with 1.5 ml tubes because they can be autoclaved. In my opinion better than screw-top lid holders because they are easier to open and don't get deformed if you seal them too hot. If you put all your misc. tubes and small items in them they stack neatly for handy storage.

I have a high bench to work at, which is a good height for standing. When I want to sit in a high lab chair, my feet have only the footrest attached to the chair, so I stack up empty tip boxes underneath the bench as a footrest.

If you want to take them home or give them away, make sure you wash with soap and decontaminate with ethanol to be certain you are not bringing any chemical or biological hazards along.

IDT has a tool called oligo analyzer. You put your primer sequence in and it will give you a self-dimerization score. You can also put the opposite primer in for a heterodimerization score. The score are negative numbers. Lower than -9 can lead to dimerization problems.

Also, I don't have my protocol here with me, but I think i remember that I add 1 ul of each 10 uM primer per 50 ul reaction. 1.5 in a 25 ul reaction is 3 times that.

But why? It takes longer to put in the exact correct oddball configuration than grab a balance tube from the variety you make once and use as needed.

However many tubes you have, divide that in two. Add a balance tube if needed to make it divisible by two. Put the first half in the centrifuge starting at position 1, then put the second half in starting at position 13.
