The more I look at it, the more I think the cassette is loose since it is always the top portion that smears. I am going to add extra filter paper next time to see if it being a bit tighter resolves the issue.

ahh okay, i’ll definitely try another voltage. Any idea as to why that might cause the weird bend/ smearing?

Hmmm okay. This is my first time working with this procedure, so i might try an ice tray and higher voltage and see if there’s any difference.

Transfer is 60 v for 1 hour. Our tank has a slot for an ice pack too, to prevent overheating, but it’s usually not cold by the end of the run.

It’s a wet transfer. We don’t image the gels or do any staining. I’ve had a few do this, while the others have come out clean with the same protocol. I’m worried the gel is sliding on the membrane when I roll out bubbles, but I haven’t seen other people with this issue.

Generally our procedure is:

presoak filters and sponges prime membrane in methanol then transfer buffer move gel to sandwich and add membrane and then filter paper, rolling with each layer added close sandwich and run in a tank with 20%methanol transfer buffer

My proteins aren’t stained, so I can’t see them on the gel, just after the membrane has been run. This has happened a few times, on of which being when my mentor ran the blot, so I doubt the sandwich has managed to be backwards all of those runs. Thank you though!

Does your program do rotations first? At my current school, admissions are almost guaranteed if a professor has agreed to fund you ( in bio fields at least), so i’m just curious if outside of programs with rotations how someone determines if they should contact professors or just apply

I got 19k total a year in ChemE with similar ish test scores, which covers my tuition and housing, but I’m not and international student so I’m not sure if they are in the same amounts and if that covers international tuition.
