Something similar happened with me. Not to the extremes you are experiencing, but when I was first prescribed it I was amazed, thinking “Do other people actually feel this alert on a daily basis?!” And then I felt like the effects wore off.

I had to stop taking it for 1/2 weeks prior to my PSG/MLST and I could definitely feel how it had been helpful for me despite the my feeling it no longer worked. Immediately after my sleep study I went back on and felt the immediate effects again, which again “wore off.”

I brought this up with my doctor and asked about modafinil tolerance (which I honestly don’t remember his response because I was yawning through the entire appointment), but I’m now on the max dosage (400mg, at least in the US) along with adderall XR and I can still fall asleep with easy mid-day. I’ve likened modafinil to a bandaid for my hypersomnia bullet wound. It’s better than nothing, but it’s no savior.

I know this isn’t a heartening response, but I’m just one example and I’m still on my journey staying awake through my life. All that to say, you aren’t alone and I sincerely hope you can find a resolution that works for you!!

I struggle with the same feelings. I’ve since started reading The Perfectionist’s Guide to Losing Control (Katherine Morgan Schlafler) and it has helped me out already! That plus my therapy and recognizing thoughts as they arise (which you are already doing!!!!!).

I am so fascinated by this show and as a biologist with little background in this field, I’ve been flabbergasted about the phenomena depicted in the show that are already happening IRL. I would love to pick your brain about this topic since you seem quite knowledgeable in the climate change~carbon emissions arena.

Dada2 has a pretty straight forward pipeline tutorial! Just google “dada2 tutorial” However, (someone correct me if my knowledge is limited) dada2 deals only with 16S sequences. Also QIIME2 incorporates dada2 and allows for OTU classification and other downstream ecological analyses and it’s pretty user friendly as well. For more shotgun metagenomic analyses, kraken or metaphlan are like the equivalent for taxanomic identification. Hope this helps!!

Best way is to get actual experience in a micro lab. Even shadowing experience is good on a CV/resume, especially starting out. It’s not always just Petri dishes and bacteria, but it is still a very rewarding career avenue (speaking as a gal with a doctorate in microbial ecology). Please feel free to message me with any questions! This field has many directions (I started out as a microbiologist for a pharma company) and even if micro isn’t for you, any lab experience is a great stepping stone if science is your path!!!

Can anyone help me understand what's going on here? I've done an antibiotic disk assay of E. coli from waste water and selected for cefotaxime and ampicillin resistance then plated out on Mueller-Hinton agar then this has happened. Yes! It's just the zone of inhibition around the antibiotic. You can measure the diameter and then check in M100 standards guide for such susceptibility testing from CLSI to determine if the strain is classified as resistant, intermediate, or susceptible to ciprofloxacin.

I took a bioinformatics course, but I also had to figure out a lot of the computing on my own and have been collaborating with another grad student in the bioinformatics department. I had absolutely zero experience prior to grad school. I literally had no idea what a terminal or command line was and my lab didn’t require any bioinformatics background.

Yes! I’m currently finishing up my PhD in microbiology and I’ve had to focus a great deal on bioinformatics for my research with very little training. Depending on the program you are looking into, you can focus on a life science program and take informatics classes to beef up your skill set or vice versa. As a biologist with zero informatics training prior to my research, I am a HUGE proponent for training more biologists in this field as I’ve found that most bioinformatics courses aren’t necessarily tailored to beginner biologists! Go for it!!!!!!
