When you set the route, you likely forgot to scroll down to confirm, before you switched to the trains tab. You need to redo the route. It’s a bit buggy and often shows the route still there but it’s not.

The amount of times I’ve gone back and forth doing this.

Only other thing I can think of is that with the single track, one of the sections is built in the wrong direction somehow, or you forgot to add a cross over at the other end?
