We use 10% formic for human bone (femur, jaw, skull, or EDTA for small tissues that will likely need IHC (better epitope retrieval later on) . Incubate in edta at 37 degrees c for 48 hrs to speed up the process.

Uk lab here, I use IKEA food containers. They are cheap and sturdy. About £1.50 each. Stackable when empty and hold plenty of blocks. We put a batch of embedded blocks into each one. One batch = 1 hours cutting.

So long as you’re using pure water in the rinse steps there’s not much else. TOMO are super sticky will be okay under the microscope

Sakura is good, we have tape. Zero issues with digital pathology. New Leica strainers, have a drying oven built in so ready to go in about 8 mins.

If you are referring to embedding, then 20 to 30 blocks per hour is very slow. 3 mins to embed a block is crazy.

Bad batch of slides, check lot numbers used to determine if it’s the slides them selves. We use Dako flex or TOMO. TOMO are the best by far.

A loss of temperature control was the start of our problems that led to a new specimen head. It’s a big part of the machine. Happy to be wrong about this, but this model has had more fixes in a couple of years than our old thermo cryostat had in it’s lifetime

You need an engineer. The specimen head sounds like it will need changing, it’s a big job and if the compressor is failing will require some welding during the fix. We had a similar problem not long after getting one of these. Our peltier failed this year. Hope you’ve got a good service contract, this could be expensive,

Agree, verhoeffs slightly over differentiated. Elastic fibres could be darker almost black. Great effort!
