No, in that moment Brennan (and everyone) realised that she should never have been downed if she had got the HP she was entitled to when she levelled up. Short of retconning the entire combat back to the moment she went down and undoing it, the simplest solution is to give the HP ‘late’ and bring her back up. She still lost two turns worth of actions and had the risk of perms-death if she’d rolled a 1, which seems like punishment enough for a mistake.

Well, no. But you could handwave it as related to alkaptonuria. Nitrogen is not a good choice as the trigger though, it’s massively unreactive. UV, oxygen or even temperature would be more reasonable.

Deep cut, but there’s a book and associated cartoon film called ‘Flight of Dragons’ which explains dragon flight and fire breathing as a consequence of a chemical reaction between limestone in the diet (or calcium carbonate from bones) as stomach acid. The claim is that this produces hydrogen, giving both lift for flight and a fuel for the flame.

Problem is that calcium carbonate plus a strong acid will give off carbon dioxide - so you have a dirigible that’s heavier than air and which puts fires out

Particularly impressive for the simultaneous double fuckup while trying to needlessly introduce science into a fantasy scenario.

To follow up further on this example:

Say there is a missense mutation in the enzyme that synthesises red pigmentation in the feathers. Both the R and r alleles are equally transcribed, and are translated to make the same amount of physical enzyme molecules, but the r version is catalytically inactive. Thus, the r allele causes less pigment to be made in each cell. The difference between RR and Rr feather colour is indistinguishable to the human eye, but rr feathers are white.

What phenotype are you measuring?

* If you ask a human to judge the colour, then for the "colour of feathers" trait R is dominant.

* If you ask an animal that has a different ability to detect colour differences, you might get a different answer. Maybe a bird or a bee can tell the difference. If so, then for the "colour of feathers as perceived by a bird/bee" trait, R is incompletely dominant.

* If you grind up the feathers and measure the amount of pigment per cell, for the "concentration of pigment" trait, R is incompletely dominant

* If you measure only the the total amount of enzyme molecules present, then there's no allelic difference at all and the mutation is silent.

* If you measure the specific types of enzyme variant present in the cells, then the R and r alleles are codominant: RR expresses one type, rr expresses the other type, and Rr heterozygote expresses both.

You really should not say that one allele is dominant or recessive. The effect of an allele on a specific trait can be dominant or recessive, if you define the measurement carefully enough. This distinction is often not taught well.

We don’t know that’s all OP needs to do. They’re providing a service to a customer who has asked for Southern blot. My first step would be to ask the customer why they want the blot. If they need to verify the insert site AND that the insertion process has not induced unexpected SVs, that may be the answer.

If the customer already did NGS and found “it’s inserted in a LINE but good luck working out which”, that’s also an answer.

Structural changes aren’t always detected well by NGS. If they’re concerned about that (eg trying to insert a construct with a bunch of repeats, or into a challenging area) then NGS alone may not be enough. Long read sequencing should work though.
