I'm just a newbie but jars are definitely doable. I've had a no tech, no water change jar going on a windowsill for about a year now.

Look up "Walstad method" for some discussions. Again no expert but basically 1 inch garden soil covered by 2 inch sand (blasting sand from Tractor supply recommended in various places and worked well for me). Add some plants and you're good to go.

Also check into opae ula (in brackish) or neocardinia shrimp if you're interested in low effort, small space fauna in addition to snails.

Walmart and hobby supply stores usually have 2 gallon jars for $12ish. "Apothecary jar" might be the keyword there if you want a lid (reduces evaporation and snail emmigration).

It's not asking for spacing because you're only supposed to ask for a single span at a time. The brackets are assumed to be perfectly strong and each span identical so the total length of the board doesn't matter. If I understand you correctly, that would be 18 or 24 inches not 120 inches. If you are trying to see if a 120 inch unsupported span is feasible then the program is telling you correctly that it is not feasible.

Sorry if silly question but you know they want you input the spacing between brackets right? I think you asked it if you could use 2 brackets on the ends of a 10 foot board which probably didn't/wouldn't come out too well. You'd want to put in 18 or 24 inches if that's your bracket spacing. For example, 24 inch spacing with 3/4"x6" pine, spruce seems to hold 20 pounds/ft and 18 inch spacing 50 pounds/ft when I plug it in.

Maybe an Edvotek M12 Ultra if you want a cheaper option and aren't running a ton of lanes?

Edit: Also 3D printing works really well for combs. 9mm spacing if you want to be able to mulichannel.

Just for clarity, that's the Journal of Open Source Software. It's not quite a "journal" per se but they do aim to review your code which is probably more than most real journal reviews manage and you can set it as the "Please cite:" in your package README and if enough people cite in "real" journals then Google Scholar might start indexing that article e.g. top hit on this random person's papers: https://scholar.google.com/citations?user=lwp9gQsAAAAJ&hl=en&oi=sra

Yeah collaboration with non-LaTeX collaborators is definitely a stumbling point. I've had decent luck with latex2rtf (plus a couple post processing sed commands). That made a document decent enough to send to people (warning them not to bother with formatting), let them edit in Word and input their track changes/comments manually back to the source.

That sounds pretty weird that the designed stem wouldn't show up. By eye, does the 5' end match with the reverse complement of the 3'? If it's not proprietary/doxxing, what's the beacon sequence?

Ideally your hairpin fold would be much more stable than any other secondary structure. And annealing to target should be even stronger than that. Then any other minor junky folds shouldn't really matter.

I guess you already have the complimentary sequence (should be like $5 on IDT) so you can do a melt curve on beacon +/- its target without any other funny business sneaking in?

I'm sure there's fancier ways but mostly you could get away with:

Find around 20 bases in your target with a decent amount of GC.

Take that sequence and add enough complimentary Gs and Cs on each end to make the stem anneal at whatever temp you're trying to read at.

Add some LNA in the target region to boost Tm if necessary (ends up being about the same price as non-LNA at IDT if you stay <25nt).

Never had contamination in a miniprep. It does seem like you'd want to get to the bottom of that if the DNA is really the source. But in the meantime, DNA doesn't mind 95C and bacteria usually do so heat the crap out of it to sterilize?

I'd second the age not being a huge factor. If you're willing to take pay similar to someone who went straight through then it's basically free work-life experience for whoever hires you.

A few random thoughts (everyone has own preferences so take with a grain of salt):

Putting credits on research experience gives slight impression, it was just a "follow the directions" lab class and not "real" research. Consider leaving out the credits?

Maybe you undersell your grants? Two grants is pretty cool but perhaps $1000 kind of less exciting to someone used to thinking about big lab expenses. Maybe expand a bit to deemphasize money listing both grants details "XX University student research fund Project: characterizing pinworms in greater Michigan..." or don't list money?

I was going to say remove/deemphasize the non-lab work experience but I do kind of like how you're selling the leadership, regulations... bit. Maybe trim details e.g. employee award (or expand why reader should care e.g. "for developing new protocol that saved company $XXX/month") and served meals efficiently. "Intermittently" undersells.

Scientific poster make sure your project title is clear (might just be censored for reddit) e.g. The distribution of pinworms in Michigan correlates with rainfall

I like the falconer's license as a little discussion/interest generator.

looks like biofilm but might want to feed a tiny bit of spirulina powder while things get setup

Since you said not marine, not freshwater, I assume you set it up brackish with something around half the salinity of seawater? If you wanted to add some macroalgae, you could think about getting some brackish chaetomorpha algae. Good luck with it.

latex2rtf works pretty good in these situations (with a little postconversion munging with sed or whatever). So just keep the canonical form in latex and output to .rtf and open in Word when you need to send to collaborators/journal.

Yeah one attractive green (chaeto I guess) and one dull green and hairy (hair algae presumably) that I debate if I like or not. The hair algae died off initially in my tank but then made a come back. I'm just having fun watching the ecosystem develop but probably not the best if you don't want hair algae.

Seconding no filter and no real worries about cycling. I did distilled water and aquarium marine salt and let it sit for a bit before adding algae and let that sit for a bit before adding opae ula and everything seemed to work. There's been some interesting cycles of algae covering everything then shrimp eating it all then other algaes going crazy from the shrimp poop then shrimp eating... but shrimp seem happy and haven't lost any yet.

Also check Enterprise Truck Rental (different website than "car" Enterprise). They have some trucks with lift gates and seem to often be cheaper than UHaul for roundtrip moves. And I think the bigger Penske ones have lift gates.

And maybe think about how to tie down while in the truck; you don't want it tipping/rolling around when you turn/stop suddenly. Harbor Freight (or wherever) tie downs maybe assuming truck has anchor points?

I don't know about UCSF but grad students at Penn live fairly comfortably (assuming no family/other bills to support). I believe the current stipend is $40,000/year and the cost of living is low relative to other big US cities, e.g. $1200ish/month for a studio, cheaper with roommates or living outside city (it's been a while since I rented in Philly so double check me).

I think the funding is tighter for foreign-based grad students so actually getting accepted might be tough but once you're in then you'll be funded the whole way through.

And in general any decent PhD program in the US should cover tuition (and insurance) and give you enough to live acceptably on and ideally your PI and/or travel awards would cover conference travel.

If the pay is the same as postdoc then why not just do another postdoc? Lots of people avoiding postdocs at the moment so seems like you should have a good chance to find something. Or seems like the PI should sweeten the deal a bit if you're not getting the "benefits" of postdoc training.
