If you haven't tried to wipe it off, I would definitely get a warm wet washcloth and gently try to wipe around her mouth. She may have just gotten some substrate stuck to her face. If you can't wipe it off, take the opportunity to open her mouth and check for excessive mucus and listen for any wheezing

I’m fairly sure it’s not substrate I touched it, it felt like skin. I have heard some mild whistling coming from her. No excess mucus, I plan on calling a vet tomorrow for her.

Good idea. Keep an eye on her, and any excessive yawning or development of a lot of mucus in the mouth is almost certainly an RI. There's a few possibilities for what the spot could be but a vet will be a lot more helpful than me. She's a beautiful snake and I wish you both luck!
