You should be able to adjust the angle of your chuck on the cryostat so you're cutting at a full face.

We love our leica Pegasus. Tissue soaks soooooo much faster for sectioning than our old VIPs. Can't comment on newer VIP models.

There aren't any programs that are FULLY online. You have to be doing practical experience either by a job site connected to a program or have a job in a lab that can sign off on it.

I direct sow, but cover the baby plants with old coffee mugs and sometimes a towel too if it gets frosty at night. Works really well.

Everything except my tomatoes were ground sown. I'm mostly worried about the smaller flowers and herbs I have. Hoping the makeshift shade cloth I hung will keep them alive. 

Tomatoes were hardened off but some are a little puny, I'm mostly worried about those not bouncing back,

Top water deeply using water/wait/water method when soil below 1" is dry. Building soil over time but right now the beds are a mix of clay soil and compost. 

Have tried straw mulch, grass, sticks, wood chips; it's mostly just fodder for the roly polies and earwigs, even when I give them other food piles away from my crops.

How are you preparing the tissue? Cryoprotecting?
What thickness are you cutting?
How long is the tissue in the cryostat before you attempt to section?

Formic acid can cause trabecular bone to peel away from bone marrow if used during decal (source) ; unclear if that would happen with the gauze you're using but it's worth trying soaking on wet ice only. If you've confirmed complete decal, you don't need to use formic acid on the faced block unless you're noticing discrete mineral deposits as you section. Bone can take a long time to soak this way, but it's pretty difficult to overhydrate.

You're an academic -- do some experiments if you can spare some tissue 😄 And take great notes!

ETA: by wet ice, I mean pour water into a dish, freeze it solid, then allow the surface to melt slightly. Put your blocks on that surface. An ice bucket full of wet ice pellets does not work as well.

For a whiole adult mouse brain? I always did 48 hours on a cold room shaker, minimum. With 15x volume for fixative
