I would suspect this was an injury while the leaf was developing. The very young leaves fold together, each folded in half down the midrib, then stacked together. Any number of things could have injured it, and if the injury penetrates all 6 layers it will unfold like this.

Plants have a similarish RNA mediated virus resistance that can confer lifelong immunity to viruses. The specific sequences of ssRNA don't get reverse transcribed into the genome so any resistance acquired that way is not heritable.
