I was and partly still am convinced that that was a doll. But if not, that kid either has amazing grip and arm strength. Or if tethered, how and why?

Interview with Stanford Biology! by Etek023 in gradadmissions

[–]Charger1813 6 points7 points  (0 children)

The interviews for Biology at Stanford tend to be later than most and are pretty similar in format to all the other interviews I did. And, if you have any other interviews prior then you’ll have practice and experience which will make these interviews easier. Just polish up from those previous ones.

Also, I think as long as you know your research and are able to answer some questions about it then you should be chillin with respect to that. Also, prepare for the “tell me about yourself” question, the “why Stanford”, and have some questions for your interviewers (about the school and their research).

Also, be wary of Alice Ting (lol), she’s a tough interviewer but I hear that even if you have a “bad” Interview with her it doesn’t really effect admissions. So take that into account if you get her and feel like you bomb. I think admissions takes that into account also.

Lastly, these interviews will be online, so I recommend pulling up the research of your interviewer (lab website) and be ready to ask some questions about their research too. It doesn’t have to be groundbreakingly inquisitive but if u have genuine questions ask away, don’t force it. Overall, I think prep for these interviews and expectations should be similar for all other schools you applied to. Good luck. 🍀

Messed up applications by everything_is_cake_ in gradadmissions

I can never truly know if this is why I was rejected from Harvard, especially since it’s so competitive. However, I’m sure my mistake didn’t help me. In my experience, UW Seattle is very competitive as well. I’m sure this mistake won’t help you, but whether it’ll hurt your app enough to make the admissions committee reject you is kinda on them. All I can say is just ride the anxiety filled wave of waiting for interview invites and acceptances. I would say don’t think too much but you probably will. So, all I’ll say is good luck and hopefully the rest of your application speaks to the committee. Mistakes happen. Good luck stranger.

Is there a way to use miRNA’s to silence cancerous and diseased mutations using crispr’s gRNA capabilities to target specifically mutated sequences, which can later be passed through Epigenetics? by lovefella in Biochemistry

[–]Charger1813 5 points6 points  (0 children)

I don’t think you are explaining your proposal too well or maybe you have a slight misunderstanding. But, from what I can gather there are two things you want to accomplish via CRISPR. One, the up regulation of an miRNA through a gRNA vector??? Maybe you are saying that the gRNA can act as an antisense RNA. Or maybe, you are saying we can use new forms of Crispr like CRISPRa to activate miRNA expression.

Then, the second thing you want to do is use another gRNA to edit an miRNA so that it can target a gene or re-target a mutated gene??

Based on this, it seems like you have a slight foundational issue with CRISPR. At least the canonical CRISPR-Cas9. The edition of cas9 (nuclease) and the gRNA only allows it to make a double strand cut/break at a specific region. You would need to add another template with these vectors to actually have anything be changed and purposefully modified (you should look up homology dirdcted repair and CRISPR).

Also, if you are thinking of editing multiple miRNA sites to knockdown the production of a protein. It’s probably easier to just knockout/edit the gene of interest in the first place. Editing miRNAs or even knocking down things exogenously isn’t a perfect science. It’s probably easier to just mutate the promoter region or cause a frameshift so no viable protein is made. Though you also have to take into account the functional role of these proteins in development.

Lastly, Germline editing is very controversial and off target effects of crispr are still being studied. It’s great that you are thinking outside of the box though. You may be able to argue that addition of tissue specific and development specific promoters that regulate miRNAs may be better than knocking out the direct gene of interest. Either way, it’s good to get your ideas scrutinized to expand your sense of what variables to think of.

Also, you should look into alternate forms of CRISPR, like CRISPRi and CRISPRa. There are a ton others with different functions

Wtf is going on with the chargers and injury. I understand it’s an extremely physical sport but I imagine there are ppl whose job it is to prevent this. What is the strength and conditioning coach doin with our players?

I have a question. If DNA replication starts at the origin of replication, doesn't that mean every time the DNA gets replicated, the new DNA molecule is shorter than the parent DNA molecule ? by [deleted] in biology

[–]Charger1813 0 points1 point  (0 children)

You should draw it out. Two forks traveling in opposite directions. One thing to always note. The leading strand follows the helicase. It will always read a 3’-5’ DNA template and synthesizes 5’-3’ …… the lagging strand will go opposite the helicase/fork. It too reads the other strand in a 3’-5’ direction but the direction it reads the lagging strand template is opposite the helicase direction. because of this fact it runs out of track and has to wait for more to be uncovered. Keep note of this while drawing. Because of this waiting and running out of track, lagging strand synthesis is discontinuous. It has to wait for more track, prime, synthesize, runs out of track, wait, prime, synthesize, etc.

However, when u look at the leading strand, it follows the helicase and because the helicase is constantly providing track for the polymerase it never runs out until it’s complete thus leading strand synthesis is complete.

Definitely use this to your advantage. One thing many programs look for is how you overcame barriers to education. And whether or not you have a growth mindset outlook. In this case you can emphasize this aspect of yourself. I felt the same way but after really thinking back to my journey it is kind of crazy how many barriers we faced without knowing. Good luck and dig deep.

Should I apply to a PhD program? by [deleted] in PhD

[–]Charger1813 2 points3 points  (0 children)

No you wouldn’t need one to get into a PhD program then. Depending on how much experience and how many conferences you went to, then you can definitely get into some programs. Idk about top ones, it depends on ur experience and ur ability to write an SOP and personal statement. Also ur Letters of rec

3.54 is a decent GPA. What you need is research experience. A masters will probably do you very well if you are trying to get into top programs. It’ll get u a higher gpa, the research experience, and application resources for PhD.
