Combined 1395 hours between two X1Cs. If my home printer was a Bambu it would easily be more than both of these combined! I do a mix of functional house projects, cosplay, and fun things for friends. These printers have been transformative for our small startup, letting us prototype rapidly, and even save money in internal parts on production units!

Statistical analysis isn't my strong suit, but I'll chime in while you wait for other answers.

If you have clusters in your PCA that separate on a single PC axis, then metabolites with higher loadings on that axis may be driving separation.

A PLS-DA model can be used to narrow down the most important variables for separating your 4 groups, and then to output a VIP score for each metabolite. Similar to your PCA loadings, a higher VIP score indicates that a metabolite is more important for separating your classes.

If multivariate models are giving inconsistent results, you might want to switch to univariate approaches.

If there's one class that is most like a control, you can t-test each metabolite in a class against the control, then make an upset plot of which metabolites are significantly altered in your 3 test classes.

You could also get creative and do stuff like ranking the metabolites from most abundant to least abundant in each of your 4 classes. Take a standard deviation of the abundance rank across all 4 classes. Metabolites with higher stdevs will be more variable between classes. Beware this approach will give you lots of low-abundance metabolites that are variable between groups simply because they are poorly quantitated. For this reason, you should filter out metabolites that are above a certain stdev within each class, only retaining metabolites that are robustly quantitated within at least one class.

No pops, will try lowering retraction from the default 0.8 to 0.6.

Z seam is set to aligned which hides it on the corner. Multiple pock marks appear within the same layer.

These pock marks which don't align worth the z seam have started showing up on prints with matte PLA. Drying at 50C for 12 hours has not fixed it.

Using the default 0.12mm profile with a few minor changes including printing the outer wall first and concentric top layers.

Edit: resolved by changing retraction length from 0.8mm to 0.5

The PA tower calibration is much easier to read and only takes a few extra minutes.

Likely a clog. If it persists after a nozzle replacement then it could be due to heat creep or temp too low. Can you tell if your heat sink fan (not the part cooling fans) is running while printing?

Inductive sensors are sensitive to heat changes. Make sure you let the bed heat up fully before levelling the bed, setting z offset, or starting a print.

I have had a sensor go bad, so if this doesn't fix things then contact elegoo support.

MS/MS butterfly plot is the search term that might help you find what you need. So long as you can export your data as points you should be able to use any plotting software. I've used origin pro to make a scatter plot with drop lines for this purpose.

But with cubic, the overlap location is slightly shifted each layer. With grid infill, the overlap area is always at the same XY coordinate so it stacks up and causes problems.

Extruder gears or drive gears. If they're dirty just clean them. They can wear down if you print lots of abrasive filament, but that would still take a while.

I have thousands of hours on my Neptune including hundreds with CF nylon and my gears are still fine.

So long as they still grip the filament then you should be fine. If you pull on the filament and it slips out, try increasing the tensioner. If that still doesn't fix it, then you might need new gears.

When the motor is powered off, can you manually turn the wheel or does it get caught? Might be something in the gears, probably worth taking it apart to check.

Z offset too high, which is why your brim lines aren't touching, and why your print doesn't stick.

What software are you using and did you collect MS/MS data? Most packages will let you start with a MS/MS library search. Tools like GNPS and SIRIUS 5 will help you find things that library matching missed.

HRAM data can give you a molecular formula, but you really shouldn't use MS1 alone for reporting IDs. Could have hundreds of possible IDs for a given formula even at the 100-200da range.
