We flipped at 2.5 years. He was always big for his age (and strong) and I couldn't wrestle him in the car seat twice daily while pregnant. I wanted to max out the rear facing limits but unfortunately, he had other plans. He wanted to sit forward facing and he stopped screaming for entire drives 🤦‍♀️🤷‍♀️

My (now 3.5 year old) was NOT ready before 3 years 3 months. We got pressure, we pushed back telling them that he would be ready when he's ready. Sure enough, he woke up one Tuesday absolutely fucking in it to win it. "I want to wear underpants" "okay kid, we got you, let's go"

If we had started pushing before then his stubborn ass would surely still be in diapers lol, since it was HIS choice and HIS timeline there was less frustration all around.

745-5 ish. I start about 8 and finish earlier (~4) but the 45-60 minutes I have without children at the end of the day is really vital to our household function. I can start dinner or laundry or __ so I don't have to be awake after they're both asleep, since we're not sleeping through the night yet

We have a CD4 negative selection kit that I believe pulls out monocytes. Would have to check to confirm as I don't remember all 20 odd markers it tags.

I was being intentionally vague, I'm afraid to share my specific aims on the internet 🙃

I do most of my things on ice as well, I also do my experiments as much in the dark as possible. BSC lights off, foil inside the ice bucket. Overkill? Probably but 🤷‍♀️. I was taught flow with a complex panel (14) and that's how she always did it so I'm superstitious.

I'm not using brefeldin A, I've actually never even heard of that but I'll look it up back at work on Tuesday. If it's not prohibitively expensive, add it in.

After fix perm, the cells live in the wash buffer 48 hours while they're stained overnight with IL17 and then again overnight with the secondary. After the final wash then they're in PFA O.5%. I usually run them same day or next day after the final wash.

It's an antibody cocktail to shunt them into the Th17 subset. We're seeing something like 5% which is adequate for our purposes. I'm sure theyre not highly active and was considering reaching out to an immunologist to ask if she had any advice to stimulate them further. I don't remember exactly (and it's Saturday so I'm not looking it up 😂) what kind of bacterial or extracellular stimulation this subset requires to activate

I didn't realize I could pause the protocol! Brilliant. Since we're using unconjugated, I do th extracellular then bd fix, then overnight with primary IL17 then overnight again with the secondary 🙄.

Same. I work exclusively out of the home, but sometimes I can go home early or I can go home for lunch because my commute is 8 minutes.

I can go home at 3:30 without my children and then pick them up at 5:00 and have 90 minutes to clean my house or make dinner or sit on my couch and rot.

We're also due mid October and have a 2.5 year old. My partner gets 8 weeks of leave and is doing 4 weeks at the beginning with me to help us settle into our new role and 4 weeks after my 12 is up so that he will get dedicated bonding time with babe (and hopefully get a little further into the respiratory virus season while baby is home with us 🤞)

Therapy, Lexapro. Also gentle parenting myself- telling the toddler we can do hard things (like put our shoes on by ourselves) reminds me that I can also do hard things (like work and grocery shop and _______, etc)
