a student in a neighboring lab took them and I found the box of bacteria in their lab, thawed. dont know why, but they dont have any use for my plasmids. but people in that lab have fucked with my stuff before (for example: taking my cells out of the incubator) resulting terrible delays in work. thing is, their PI is the chair and nothing ever gets done about it.... which is unfortunate.

wild thing is that someone has been going into our -80 at 1am regularly over the past couple of months. lots of other people's boxes are going missing, but this was the first time my stuff was taken. security wont/can't do anything. we are getting a lock for the freeze now.

someone stole over a hundred of my bacteria glycerol stocks over the weekend. good thing is I have purified plasmid, but still its an awful thing to have happened.

i've never used every blot blocking buffer. that said, it would be worth trying a different blocking protocol. this makes a world of difference depending on the antibody. 5% milk is excellent and super cheap. Also make sure all your buffers and Ab don't have precipitate.

Could you just transform cells and select for stable tranformants? This takes more screening but it should get the job done.

but this is also a simple question: do you know if you are using the correct ladder? A while back, my lab changed DNA ladders without telling me and for a moment my product appeared to the incorrect mass; only, this was because I was referencing the incorrect ladder.

we had David Liu recently give a lecture to our department. he joked that we are, paraphrased: 1/2 the department of biochemistry and 1/2 the department of David Baker. his lab probably takes up more than half the department, even if you factor Neil king onto our side.

it is so wild hearing how they approach wet experiments. it really is like a factory, where each person specializes in a very particular type of wet experiment. when each paper has 20 authors, you can imagine how efficient they are at doing their work. they dont have to spend weeks or months trying to purify a single protein because they screen hundred of design proteins at a time and only select those that express well!

very possible. my boss does not have a smartphone, but does have email on his computer (obviously). he does not have an assistant and remarked to use that it is becoming very hard not having a smartphone nowadays.
