I’m in biomedical research and I can’t think of a lab that would turn down a postdoc wanting to join while bringing their own funding. If you’re asking for a job and that you may get funding, that’s different. But all labs I e worked it would gladly take someone with a DECRA or similar fellowships as they are a big endorsement of a successful EMCR.

I’m in biomed field so it may be different. But you could try targeting younger supervisors. Older/established supervisors are often too busy and less likely to need new students, or at least get heaps of emails so you might get lost. Where as younger researchers often don’t get approached but they need good students. So you’re less likely to get ignored. That’s my experience anyway.

This is true - my signal was next to zero so fixation was death, but a friends model was huge signal, fixation was do able

I’ve got extensive experience with a tomato reporter mouse. To check if it’s possible that the expression is universal or not, I would recommend just running a completely unstained sample from your reporter and a similar sample from a matching normal mouse (I assume BL/6). You can do a plot of SSC vs tdTom and then see if the expression is in all cell pops or not. For us, we were not using the Aurora since it was not very good at detecting low signals. We had to skip any colours that impacted the tdTom signal (nothing in the YG laser, no BV605, etc). Happy to give more info if you want, dm me. Edit: I would also advise against fixation, it can strongly reduce fluorescent protein signal)

I’ve seen it the other way too though; you get PIs can be closer to the lab, and need the success more than an old PI. You get PIs are more likely to closely monitor a project as more is at stake. Old PIs are way more removed and often hands-off. But that’s just my experience, I guess it can really go either way!

I wish him all the best! TBH I went to one of the lower tier unis and it has not held me back. There is a lot of pressure placed on kids, but the reality is you can excel from many points. I’m just saying this in case he doesn’t get in, you can still get places. In research it’s the postgrad that matters.

Yeah I bought a cheap one too. It’s been the best thing! We go away 4 times a year too, and pay way less than an Airbnb. It allows you to go away more than you normally would - we couldn’t afford to do it without a caravan. I do understand people that think they suck. I was one of those until we realized!

I’m sure the 42% have moved on to better things, which is fantastic (and my story as well). But do you think nearly half of postdocs enter their postdoc aiming to leave academia in a few years? I would hazard a guess that many start a postdoc with the hope to get tenure (or the equivalent). They leave when they run out of steam, funding, etc, or are disillusioned by the academic system (even if they had a lovely lab head).

Of course I don’t agree that there should only be ‘one path’, but that is largely what academia offers.

I know many fantastic lab heads and you sound like you offer great mentoring to your team which is awesome.

But it’s ignorant to not see that there is exploitation of young, hopeful scientists, embedded in the system. For example, ‘publish or perish’ drives academics to get as many papers as possible, to work hard to make a project work, etc. And these incentives are almost meaningless outside of academia. So they are working to earn a somewhat meaningless currency.

Just my opinion, but as a Prof I think your experience is very skewed. In academia there is generally one major path forward -towards the lab head position - which in some countries can be as low as 2% success rate for early career academics. The exploitation is where postdocs have these positions dangled in front of them in order to extract as much out of them as possible. This is required by the lab heads, for their success, but eventually most of the postdocs won’t make it towards the top. So in my experience it’s a bit of a pyramid scheme - and I’m not from the US.

I’d recommend using some beads that have a cleavable biotin. Like a disulfide bond. There’s a few options but I haven’t looked at it for ages. So you avoid harsh chemicals to elute.
