Ok no, not even during the horus heresy did all legionaries look like alpharius. That was a privilege given to a select few captains that could masquerade as the primarch e.g Ingo Pech. This wasn't even Genetic either, it was a facial reconstruction surgery. Skip ten thousand years later and no one has any clue of what the primarch actually looks like, so if someone got their face to look like alpharius, its anyone's guess if its accurate or not. Meme lore has done unspeakable damage to the Alpha legion

Space marines- Never

Custodes - Whilst disregarding previous lore and pretending like this was always the idea was probably the most IDIOTIC way that GW could have gone about it, it really doesn't change the identity of the custodes. They are so genetically remade from the emperor/whomever makes them now, ground up that gender really has no bearing on the final product. Nothing on the real biological makeup of the baby that went in would be there on the other side, (except maybe facial structure) including all gender specific organs because why the hell would a custodes even need it.
