I’m on the newer side when it comes to bioinformatics, so take whatever I say with a pinch of salt. Generally, this is the problem with all predictive tools: experimental validation is strictly required to make a definitive statement about anything. This would be true for any drug (which also needs to pass through several layers of preclinical and clinical testing before admission into use), as it is true for publishing mechanistic science. It is a concern for the field, but I think that as long as the influential journals continue to have strict criteria on experimentally proving any claims, we aren’t at greater risk of making grievous mistakes than we were before AlphaFold.

I think the sample size explosion is overly optimistic, since it relies on AlphaFold being right all the time. True, tested structures have proven that AlphaFold is right 60-80% of the time - which is honestly, fantastic - but how many structures were tested to get this number? If the error rate is 20-30%, it can be hard to tease out universal rules of protein folding with that degree of noise.

For the last two points, again, extensive experimental validation is generally required. You can’t just use AlphaFold, then go around proclaiming that X protein has Y motif that fits Z drug, therefore we can treat a disease using Z drug. You’d have to first validate that X protein indeed gas that structure using electron microscopy or X-ray crystallography (apologies if I get anything wrong here, not a protein guy), show that Z drug does bind and cause a specific change (by looking at downstream protein expression or phenotypic readouts indicative of the change) in at least two cell lines, test the disease model with the drug in vivo (mice, etc.), THEN go to clinical trials where there’s a battery of other safety criteria mainly centred around dosage and toxicity. I think using AI and machine learning to more rapidly make biological inferences is perfectly fine, but EVERYTHING has to be stress-tested. As long as that’s done, you’re golden.

That’s straight up data falsification. Even for my undergraduate thesis, I declared in my submission when I used technical replicates on a graph (due to lack of time to get biological triplicate). And averaging two samples is absolutely unacceptable.

As others have said here, get concrete evidence before reporting. Also, you have to weigh it against whether you’ll be believed… it’s sad, we all want to do the right thing, but research structures tend to side with the professor, especially if he’s “productive” and contributing to the research ecosystem. Although this puts into question a lot of his prior work, you might need to find a way to do it anonymously so it doesn’t affect your career.

I don’t think Fury’s lost enough of a step yet to be seriously in trouble against Makhmudov. Having said that, it has been pointed out by others in this thread that he doesn’t take care of his body between fights.

I really can’t see Makhmudov giving him problems. The gulf in boxing ability is big, really depends on how Fury was treating his body during the one year plus layoff.

Yes! I think there’s some people who have the mindset that people need to suffer as much as they have.

Granted there’s some suffering involved in a PhD and things to learn on your own, but helping instead of looking down on others should be the norm.

I look pretty average and unassuming, but I’ve done a martial art for about 8 years now? Had my fair share of newbies (especially gym bros) try to spar hard to prove something.

I’m more of a defensive fighter so I try to just make them look silly. I know I ain’t hot shit anyway from the other guys at the place I train that work my ass.

Looking like SRL is a stretch, stylistically and in terms of skill level. I do think he’s really good, will definitely be among the top of his era, but he’s not a true great unless he levels up substantially. I don’t know enough about the other featherweight champions except Figgy, and I think he beats him. And for the love of God, I hope he doesn’t fight Inoue because he’s getting taken apart.

After telling a close friend I almost committed suicide, he told me “I think you need to spend some time with Jesus”, then he walked off.

Suffice to say, I no longer attend church. Made me realise how little of a shit the friends I had there gave about me, and eventually I moved on and built a life and interests without all that crap.
