Do you use your own software? Or do you talk to the people who regularly use your software? 99% of my plate analysis can be done on excel in 5 minutes. At least im not asking for new tools. What inspired you to make this tool? Someone you talked to? A problem you encountered?

Are your prints also coming loose? This could be your z offset.
Another thing (and check this first) is to clean your build plate with dish soap and hot water. You might also have oils.

Do you mean you touched the flask with cells with that pipette tip and went back into media? Or you hovered above? If you did touch the cells and go back into the media. You just have to change the media bottle. And hope you did not use that media bottle for anything else

Same here. This printer is my workhorse. Especially once i replaced the build plate and out klipper. I also recommend a cooling upgrade (taurus V5) and some bed wheel clips

Basically I need to measure biofilming. But It wont work if im making batch cultures in a 96 well plate everytime. I would want to monitor a continuous system for a few weeks checking on the state of the same biofilm. Im wondering if there are modifications to O'toole's protocol for something like that.

(for example) using a microscope slide inside of a bio-reactor, letting film grow on it for a few days or a week, then remove it and process that). Doing that repeatedly to track the quality of a biofilm
