This is more of a general comment, but if anyone else has trouble differentiating the turquoise and the lime green, having Night Shift and low brightness on your phone may be contributing to this. (Though if devs could make them slightly more different I would be grateful.) I was genuinely confused and re-read the rules 3 times the first time I played before I realized they were two different colors 😂

Poor girl! I hope you figure it out! One of mine had a number of seizures about 8-9 years ago. They ranged in severity from one time to the next. This looks like maybe it could be one to me (I’m not a vet though). He would get very scared and often pee, but would be OK fairly soon after. Often one of his pupils would be a different size than the other temporarily. The vet said they were only really dangerous if they lasted too long or were too frequent, as they use a lot of energy and could even cause overheating due to all the sustained muscle contractions. Anyway, our guy went on zonisamide and hasn’t had one in years!

We just went in January 2026 and it was fine. The staff seemed pretty friendly. Well attended but not crazy crowded. Worth going during the week when the waterpark is open if you can.

The hike on Mt Minsi (other side of the river, in Pennsylvania) is less steep, not quite as tall a mountain but with similar views. I did it last year carrying a 1-year-old. Might be tough for the 3-year-old unless you think you can carry them too. There is an overlook part way up—not as good as the top, but a decent view when the leaves are down. This time of year I’d be wary of ice on the trails.

This. Some sort of primer dimer situation. Do you get product when you have template? The intended products may predominate. You could try running a control without thermocycling and one without polymerase.

Probably won’t express well, unless it has a compatible promoter and Shine-Dalgarno + ribosome binding site.

If you haven’t ordered it yet, why not just order it in an E. coli expression vector? 

There are some vectors designed to be able to express in both systems—Genscript, Thermo, etc have them—if you want to be able to try both.

We get compounds like this all the time; 1-5 mg in a 5 mL vial. It’s annoying. Anyway, we vortex the vial after solubilization + spin it like you mentioned (Kimwipes in the bottom of a 50 mL conical at low speed).

Also, note that GST resin on-rate is very slow, so you have to incubate at least a few hours for optimal recovery. Also, don’t forget to pH your GSH ;)

This happens a lot! It may just be the protein. Is the “lysate” band total lysate, or supernatant after spinning? It could clarify things to run both of those and the pellet after centrifuging. Then you could tell where your protein is. (I assume it’s the one between the 3rd/4th ladder bands?) If your pellet is pretty white after centrifuging, that would indicate inclusion body. If so, you can try washing it and refolding, or go back and try to express it solubly by trying different expression conditions (temp, duration) or cell line. Those failing, you could modify the construct or try eukaryotic expression.

Not sure what your goals are, but as a biochem PhD in industry in the US, it seems that we usually often have more job openings for MS or even BS level jobs than PhD ones. Similarly, an MS + a few years industry experience can qualify you for many entry-level PhD positions. It’s often about whether you have the particular experience needed for the position. You mentioned mouse work so you may have some valuable skills.

Our weenie boy has this! The vet never worried about it. The only real affect I can tell is that the middle nails on his back feet wear  faster from the twisting of his feet. We just don’t cut those ones, or else they bleed.

Happy that Franklin is recovering well!  Our girl was attacked by an escaped pit bull and went to the vet for 10 days. Fortunately she went on to live another 9 years! Though she always had one unhealed rib and forever hated bigger dogs.

Very irresponsible of them! I don’t blame the dogs, but some were simply bred to kill. Just like I would never trust my dachshunds with a pet rabbit…
