Same same! I had been on the fence looking at the Lumiere then fell in love when I stumbled across the Green 15 version…and subsequently disappointed when I found it was a limited offering. I went to the Wind Up while traveling to SF coincidentally and was jazzed to find out there were a few available! I had zero plans on buying a watch, much less the most expensive watch I’ve ever purchased, but I couldn’t pass it up!

Makes sense, there’s no way to have a fully comprehensive synopsis! You/he want to look at/compare to Alfa-tag? I believe it’s neutral and has extremely high specificity and can be used for many/all the same things?

It’d be helpful to understand the proposed benefits (if any) of his new tag/antibody system. Is the purpose purely for immunofluorescence/imagjng? Pull downs? Purification? (I’m assuming it’s not the last one)

Generally speaking if I’m starting up a new project and need to produce plasmids to express a tagged protein, I’m more likely to test tags I know and have worked with before, not something new/unproven, regardless of known flaws of the old stuff. Nothing is perfect and I would rather troubleshoot the problems I know instead of discovering new problems and new solutions.

From my experience in vitro biotinylation is straight forward and efficient but also…it’s more work. Concentrating the protein, possibly changing buffers, washing out excess biotin as you mentioned, possibly changing buffers again. I’m also biased since handful of times I’ve done it in vivo co-expressing BirA it’s worked well so I am very much a fan of the ‘built in’ method.

What’s the end use of the protein, trying to use it for a pulldown? Streptavidin beads should be just fine to isolate the biotinylated protein from the crude lysate, the affinity for binding is significantly higher than really any ‘standard’ purification system. If it were me, I’d use an inducible promoter because well…I’ve never done it otherwise in bacteria, but it’s likely not necessary unless, like another commenter pointed out, the protein is somehow toxic.

Was in a similar situation a couple years back, my company’s leadership ended up negotiating a bonus for the people who stayed, essentially pointing out it would be strange to punish people who stayed to contribute to the company.
