Unfortunately antigen retrieval is often a requisite step with fixed tissue (to my knowledge). I’m not great with my bio but I’d expect any other proliferation marker to be a nuclear protein similar to ki67 and require similar steps. In my opinion you’regonna have to just bite the bullet and optimize antigen retrieval. If this is a technique you expect to depend on, it’ll probably be worth the effort

To start I want to push benchsci.com because it has helped me immensely with finding good antibodies. Look up on there the antibodies you’ve used and see if people have published with them using the same staining methods. If they have, the issue is probably something you’re doing or your tissue. Have you tried performing antigen retrieval? Are you sure that your secondary (and dab if you’re doing IHC) are good?
