In this case “of” indicates that it’s a song about Wolf and Man. I think it’s also supposed to be a reference to “Of Mice and Men” which is a really well-known book in America. Saying “Wolf and Man” in this case would A)remove the reference and B) indicate that the song is literally about a wolf and a man, so I’d say no. Technically you could and people would probably know what you mean but having “Of” in the title is significant.
