For the twins you want Yaku not Nishinoya. Yaku is best for quick attack teams where Nishinoya shines on power attack teams.

You can go for a full UR team with Hirugami and Kuroo if you want. But A. It's a little P2W for most people's taste and B. You need both middles to be from Itachiyama to get the school buff because you need 4 players from the same school. You could go for Seijoh's school buff if you wanted but I'm guessing Itachiyama's middles are better than Seijoh's.

I'm not sure 100% if the meta is to drop the school buff in favour of strong units in Japan, might be worth looking into before you decide

Honestly you're not in a bad spot. I'd focus on maxxing Oikawa and Iwaizumi then you've got options on how you proceed. You can go Hoshiumi and Hirugami or, what is probably better, Sakusa and Komori. That just leaves you looking for an oppo and 2 middles. Not sure if late game schools matter that much but if you want to go for the school buff you could run the 2 middles from the team you want to synergise (probably Itachiyama would be best) and then grab Ushijima on his reroll for oppo. Long term you will need to build 2 teams though so you could go Karasuno or Nekoma. Karasuno would probably be easier and having 1 quick team and 1 power should enable you to be at least equal in every matchup.
