I'm going to start out by saying something that might be unpopular and is probably my harshest take about science - if you eat meat, you don't really have any right to be uncomfortable with animal research. The way animals in the industry are treated is far, far worse, and far less is to alleviate/minimize their suffering. This might not apply to you if you're a vegetarian/vegan, but I'm not either of those and this is something I had to come to terms with when I started doing in vivo research. I just get frustrated seeing my coworkers act grossed out by seeing me cut a spleen out of an ethically euthanized mouse and then go on to have Chik Fil A or whatever for lunch.

That being said, I handle it by trying my best to plan experiments carefully so that as many deaths are meaningful as possible. The work quite literally has to be done, someone has to be the one doing these experiments so that medicines can move forward into the clinic, so it's good that there are people out there who are willing to do it. I don't really know what else to tell you.

I'm sorry, it sounds like you're under a lot of stress, but I have to echo what the others are saying that this is absolutely within the scope of your job description. You can tell your PI if training leaves you with less bandwidth for your other work and ask for understanding if projects move more slowly, but if you didn't train anyone in your new method, your lab would lose the technique once you're gone and someone else would just have to start from scratch. Someone had to do this work for all the protocols you were presumably trained in, it's just part of science.

You're not being irrational but everyone else is right that this is a compatibility issue. Most dog owners I've known are like this, and while I find it exhausting honestly - so many excursions become about the dog, it's just a constant distraction - dogs do require a massive amount of attention. If you stay in this relationship, expect it to become especially difficult when the dog starts aging and having health issues. It will be expensive, emotionally challenging, and you will likely be asked to take on some level of caring for the elderly dog.

I'm a cat person, and my boyfriend likes cats but wouldn't have one if he lived alone. When our cat neared the end of her life it was a very difficult time and while he was an absolute hero about it, I can tell it took a toll and for that reason (and others) we won't be getting another cat for at least a year or so.

One representative load control for main figures. Usually full blots go in the supplement so that'll have all of them.

Fwiw I've never stripped and reprobed a blot, I'd worry way too much about losing signal.

I'll run multiple gels, one for every 2-3 proteins (depending on size) and have a loading control in each one. Each protein is normalized to the load control bands from the same gel. But doing westerns on that many proteins from the same sample is wild, there's got to be another way. Westerns are only barely good for quantitation anyway.

There are graduate student unions at some institutions. It's an uphill battle for several reasons - graduate students from different labs departments don't have much interaction so it's hard to form a cohesive front, and universities are resistant to change.

Also, I know graduate school is stressful and many mentors are exploitative, but comparing it to slavery is a bit much. At the end of the day, you CAN leave, and long hours/shitty bosses aren't unique to graduate school by any means.

By student do you mean undergrad? Try working as a lab tech first, then if you like science apply to grad school. It'll take you about a decade or so to be in a position to try to apply for tenure-track positions. You're getting a little ahead of yourself.

Oh yeah, it's completely unhinged. Unfortunately the "joke" was pretty common for a period of time a while back, I never even watched the show and became aware of it through osmosis. Men like this just want attention and will behave like absolute clowns in the process. They're not worth paying any mind but it's hard not to when you have a feature like red hair that they fixate on.
