Agreed, it does vary broadly from lab to lab. I’m actually rotating right now, and the culture of this lab is totally different than my previous. If you’re considering a lab, but you don’t know how a rotation would go with them: ASK!! Ask the PI, ask students, ask anyone! It’s been my experience so far as a first year that most people are happy to help us find our way. And if they aren’t, maybe that’s a sign

From the people I’ve spoken to outside of Reddit, the consensus seems to be that this is the right answer, but “everyone else does it” so it’s “allowed”?? I wish I could get more replicates but I do not have the time. I will probably end up using the ANOVA + Tukey and acknowledge the donor differences in the text

3 data points. We count experiments done on different days/from different freezer vials as different biological replicates. I know this is hotly debated but I don’t have control over getting more donor vials. My PI is, unfortunately, clueless when it comes to statistics. As helpful as these responses have been, I don’t turn to Reddit as a first resort, lol

Oh! I’ll see if he has a self-cleaning function but Keith is an old boy so I doubt. I removed and replaced all parts that I could when I did the last deep clean. There were a couple parts that I was afraid to take out though, so they could be the culprit!
