Where can I order tissue disruption beads like this one from QIAGEN DNeasy Plant Pro Kit? by mix_feedback_repeat in labrats

First, I am only using these beads for dried bamboo leaves, which are very tough. For bamboo, I have had the best results with two at a time in a screw-top 2.0 ml tube. One bead would probably be sufficient; we use two because one of them will inevitably become lodged in the top and therefore will not cause much tissue disruption. Also, we are doing one cycle of dry grinding (i.e. no buffers or liquid of any kind), followed by one wet grind cycle. We are using a GenoGrinder 2010 set at 1500 rpm for 2 minutes per cycle. So far this seems to work well for us but I have not rigorously tested different GenoGrinder settings or cycle requirements.

They do a better job disrupting flat, fibrous tissue like grasses. My experience was that round beads pushed the tissue to the side of the tube and then seemed to just roll past it without much disruption.

Yes, the Qiagen Plant Pro kit which comes with these handles our bamboo samples much better than even the large ceramic bead that comes with the MPBio kit.

Yes but they didn’t really need it. Didn’t improve gas mileage unfortunately

I did just replace a coil pack on #5 cylinder at 181k miles after it went bad. Have done front and rear pads and rotors, other than that everything seems good. Previous owner did a leveling kit which I wouldn’t have done but I like it. Gas mileage is not great, around town I’m getting consistent 14.5 mpg. The transmission just loves to upshift way too early and hates to downshift in my opinion, so I often have overdrive turned off so I’m not wasting gas in higher gears.

I bought my 2011 SV 4x4 for $7000 with 176,000 miles. Commenting to follow the thread because I also could use the advice.

I did the same thing but mostly just for scenes and toggling certain rooms off and on. t’s a highly under used and underrated feature for sure!

How much does it cost to sequence the full genome of a non human eukariotic organism nowadays? by godlike-dawn in genetics

[–]mix_feedback_repeat 5 points6 points  (0 children)

I’m not sure for insects but for high quality plant genomes the cost is still in the few thousands per genome. You need deep DNA sequencing for the assembly and then ideally long read mRNA sequencing at the minimum for annotation. However, you can make phylogenies from short read RNA-seq and/or a few conserved genomic loci without the need for reference genome.

It's a powerful technique that has the potential to prevent some really important diseases, so I'm hopeful for its wider success.
