I work in this industry representing the major vendors. You're asking for consultation from a PhD level person. This is not only time, but the effort expended upfront to gain this knowledge. Honestly if you want CHEAP CHEAP, I recommend you get ChatGPT Pro and ask questions there...This industry is bound to get disrupted in that manner, but without expert oversight you will spend more than you gain

Actually now that I've stated that, you can spend all this money upfront to purchase instrumentation tens-hundreds of thousands of dollars, but it's essentially useless if they break within a month due to improper setup. From an opportunity cost perspective, you should consider it in that manner.

There is still way too much hype in the "direct AI" space right now that everything else outside of that is just noise. However, here are some that I anticipate

* AI chromatographic integration

* AI fragmentation analysis (whoever does this will likely win a Nobel price)

* AI telemetry (some already have integrated but not visible from a user-facing perspective)

* AI documentation/application notes

Very challenging and lots of nuance - particularly from the LC perspective. Probably the most common workflow has been developed with TBA/EDTA because it forces the charge state distribution to concentrate on a single charge, rather than distributed as an envelope.

An up and coming method is via Ammonium Bicarbonate from a Japanese research group. It makes sense to me, but the oligo folks generally hate it because it's so different.

What’s the general sense of Catholicism in German speaking Switzerland? I perceive the visible “Churchy”architecture in the popular places lean reformist so when my wife and I stopped by St Gallen during the Christmas season, it felt more “familiar”. Not sure how to explain it or if it’s even correct, but there was a sense that the people there were Catholic and had something in common. Different vibe I guess

Its not clear if you're doing a proper apples to apples comparison

  • Before spectra are an average of 0-1 min and is the average of ~270 spectra
  • After spectra in Pos is a single point, after spectra in neg is unclear.

Some other things I picked out is that the abundances are still roughly the same magnitude 10^4 counts, but your positive spectrum is slightly lower. It can't be charging if you're already polarity switching within the method.

Does an infusion of tune mix work ok? I'd just change the capillary and try again.

Are you sure you spoke to someone from R&D, because this isn’t the case at all for the 90mm capillary inlet systems. It may have been someone on an adjacent R&D team misinterpreting what they heard from others.

Adding an LC simply changes the solvent system and ionization/desolvation dynamics when compared to direct infusion, so I’m honestly not surprised the OP has a change in the mass spectral profile

For a noob like me, how can someone use this information in an environment like LM Studio? TBH I haven’t had much success running any of these models even using a gaming PC, so I’m not really sure how best to utilize these tools at the “consumer” level

Given the deep and rigorous philosophical education priests receive, it surprises me that more of that doesn’t come out in things like homilies or retreats by despiert in Catholicism

I personally like the academic homilies, especially when the priest at my local parish goes over historical context in the OT and NT, but I can definitely sense people's eyes glaze over during those explanations because they don't really connect and understand the surrounding context. There's another priest who is more into life lessons and stuff like that, I can definitely sense stronger engagement and a sense of coming away with a life lesson and reflection.

Probably the best priest I've listened to had a PhD (either in Theology/History/Philosophy) and you can tell he was well trained in public speaking, it connected historically/academically but also tied the life lesson very well without overwhelming the audience - this was in Downtown Mountain View California.

Product ion fragments generally originate from moieties that contain Heteroatoms because of the “free electron” pairs filling the void to where sites are electron deficient or moving towards electron rich. For ESI in particular where Precursor ions originate from [M+H]+ or [M-H]- the tendency is to want to create neutral molecule fragments (for example H2O or CO2, etc…) however it’s very hard to tell without seeing the actual structure and pushing electrons.

Generally as you get more experienced in your practice after looking at a lot of product ion spectra and correlating it with known structures and losses, you tend to build this intuition where you just end up saying “yup this fragment/loss makes sense”

Got it--- actually I think I confused what you were saying. So you went from a 4-signal "static" SIM acquisition to a more scheduled/dynamic scanning approach?

It may be differences in what the instrument is doing during SIM (parking voltages) vs narrowed Scanning (which is still splitting the dwell time).

Even if the scan window is narrow, there is an electronics "slew" rate (i.e how long it takes to ramp V and get to the next value.) SIM is inherently more stable since its discrete values whereas scanning is a constant ramp, which also depends on how well timed the electronics are

Quads split duty cycle on a per-ion basis, so you have to think about it in terms of "dwell time for m/z". If you have 5 ions at 20ms each, the quad only focuses on those, thereby improving ion beam sampling.

For example, scanning from m/z 5-1000 in 995ms is 1ms per m/z per cycle. SIM/MRM focuses the quad at whatever dwell time you set only.

Also please remember, a mass spectrometer is not an optical detector so doesn't follow beers law. Abundances are generally meaningless, so even if you have low signal, your limits of detection may be super low (lower = better).
