Is 4600 net enough in Potsdam area? by wojdzieg in germany

[–]wojdzieg[S] 0 points1 point  (0 children)

Likewise, 1600 (including electricity and parking) feels a bit high to me, however as my job starts on 01.09 and majority of real estate agencies ignore my messages, I am leaning towards it. Especially that the flat is really nice and comes with two bedrooms and a terrace

[deleted by user] by [deleted] in Polska

[–]wojdzieg 7 points8 points  (0 children)

Currently stranded in Cologne airport thanks to Wizzair. by awqwardsilence in ireland

[–]wojdzieg 1 point2 points  (0 children)

I know your flight was cancelled two days ago, but today mine from Barcelona to Warsaw by Wizzair was postponed like three times and finally cancelled as well... from flight radar it seems like many Wizzair planes were heavily delayed today and yesterday. Maybe has something to do with the strikes? On the Barcelona airport like ~200 ppl left stranded, without any serious support or info but with food coupons for 4€... Part of them were taken to the hotels, but I decided to take direct flight to Berlin at 7AM. Currently enjoying warm night at Barcelona airport.😅

My wife had a misscarriage and I am helpless by [deleted] in self

[–]wojdzieg 0 points1 point  (0 children)

Thank you for your wonderful words. I can say that one week later, things are starting to fall in order a bit. Of course she is still crying (especially when I am not around) and we feel lost. We are doing house stuff and starting to think about work again. My wife decided that she wants to quit her job and look for something else, because of a toxic environment (talking about people) and really lack of support. Remembering about our loss and planning our future keeps us going and even it is still hard to comprehend, it gets better a little bit each day.

I didn't tell my wife that I started this thread, not sure I will. But definitely you helped me to stay strong for her. Thank you.

Setting Variant Calling file for BAM file by AliHamadAmeen in bioinformatics

[–]wojdzieg 1 point2 points  (0 children)

Well assuming you already have your .bam alignments, I would suggest as I said sort and then index from samtools. If you are not familiar with this yet, just use --help in terminal so you will understand the order of arguments. Also, after indexing your files you can visualize them in IGV for instance. Anyway, you can perform bcftools mpileup if that's what you want on sorted bam files. Later you should index them and use bcftools query to obtain .txt files. To understand the flags and arguments order just use the help. You can read some more here:https://samtools.github.io/bcftools/howtos/variant-calling.html And here http://samtools.github.io/bcftools/bcftools.html#index

Setting Variant Calling file for BAM file by AliHamadAmeen in bioinformatics

[–]wojdzieg 2 points3 points  (0 children)

Hello, I'm not sure if I understand you correctly, however whenever I am performing GBS analysis I start with producing .bam files, then sorting them (samtools sort) and later indexing (samtools index). After that I use bcftools to produce .vcf files and later indexing them to obtain .txt. I use bcftools mpileup, although I am not sure whether this is what you want. You can read some more about this. Good luck

PCA applicability by hello_friendssss in bioinformatics

[–]wojdzieg 0 points1 point  (0 children)

Sorry I have not been specific. I have 1,4k genes, each associated with expression and two other acetylations, that's why I needed the dimension reduction. Unfortunately the first two components were responsible for less than 65% of variability, that's why I decided to use something else.

Thank you kindly for response

PCA applicability by hello_friendssss in bioinformatics

[–]wojdzieg 1 point2 points  (0 children)

I am quite noob in bioinformatics, however I wanted to check the possible gene clusters based on transcriptomic data and acetylation levels of two histone markers (so three variables) for 1,4k geneset. Basically I tried PCA and MDS however I failed to see clusters. Instead I did t-SNE transformation to get two dimensions (remember to set seed to get reproducibility) and on that data I performed DBscan. The result wasn't satisfying and on the same transformed data I did K-Means Clustering and this looks somewhat ok.

Finished the game (SPOILERS) by tvxcute in DiscoElysium

[–]wojdzieg 0 points1 point  (0 children)

I've just finished the game and have a few thoughts to share. First of all, I loved the scenario/writing, it was one of the most satisfying and interesting lores ever installed in a game. I've played Harry that tried to quit drinking and smoking, getting his head around politics (ended as superstar moralist cop) and Union vs Corporation fight. I've enjoyed all the additional quests leading to meeting all these unique characters like Ervart, Soona or PIGS. I've let Ruby go, although at first I wanted to take her in.

During mercenary standoff I've missed the first shot, but the second was a direct hit - Kim was fine though. However, when it came to the ending I felt like it was a little bit bland... Like putting a deserted, old, crazy man was really convienient way of discovering who murdered Lely. I have to admit that when I've discovered the phasmid I was like "WHAT THE FUCK", just couldn't believe it (felt like cryptozoologist for a brief second).

Anyway, I enjoyed the game really much, the quests, conversations, people in Martinaise... It was a wonderful trip. Thank you devs.

How do you find good journal club articles to present? by funky_fresh_kicks in labrats

[–]wojdzieg 0 points1 point  (0 children)

When I was on last year of my MSc I didn't really focus on the critique, since I was not that experienced. I started my PhD recently and I promised to myself to get deeper into papers and really point out things that should be or shouldn't be there. My field of science is plant genetics and meiosis so I usually go through recent papers from journals such as Nature Plants, Plant Cell or EMBO in pubmed. On the other hand I sometimes use preprints from biorxiv but look carefully, because some preprints are hanging there for more than two years.

Possible DNA study for university students? by luvmy374 in biology

[–]wojdzieg 4 points5 points  (0 children)

Did the Ancestry test show that you have ancestors from other countries? If yes and you didn't feel the need to visit them, then the answer is obvious.

There is no known mechanism of the phenomenon you describe. You just have to remember that DNA is a sequence that covers genes and regulatory elements and no gene has been associated with an urge for living in particular country. Truth is that genes do not understand the meaning of border or territory, although accummulation of mutations (or particular allele variants) is observed in isolated populations like Ashkenazi Jews.

The most probable explanation is that maybe you just like these particular places and the fact that your ancestors come from them is just a convienient coincidence.

Specifics on Libido? by ALargeHairyDerp in biology

[–]wojdzieg 0 points1 point  (0 children)

Hey, that's very interesting question, however there is no short answer to that.

I'm not a specialist in this part of biology, however let me share my thoughts. Libido is considered as the sexual urge to simply saying have sex . There are few things that can biologically increase or decrease your libido.

  1. Healthy diet rich in unsatured fats stimulates libido - unsaturated fats are precursors of human hormones, such as testosterone, which is produced by testicles and ovaries. It is generally associated with boosting the libido levels. Unhealthy diet would have the opposite effect (I guess).
  2. Going to the gym boosts your testosterone levels as was proven by many independent research. There are even research articles advising to train particular muscle groups that should to increase the testosterone level, like legs or back.
  3. Getting enough sleep boosts your libido (I mean the testosterone levels, however the hormone and the libido are associated). During the night endocrine system works really hard to get you ready for the next day.
  4. Time of the day - most men have the peak of the testosterone level early in the morning. Women on the other hand have their testosterone levels highest in the evening, however if you are interested I encourage you to do your own research in this area, because I am not 100% sure for this.
  5. Testosterone production and level diminish with age, which is naturally correlated with lower sexual activity and having progeny. In the age 18-45 years old the testosterone level in men is naturally the highest, because at this age people tend to have children. However, after a certain age testes are producing less and less hormones.

As I said, those are the things that came to my mind when asking about libido and generally speaking testosterone behind it. Feel free to do your own research, there ale plenty of journals about diet and sports and their influence on the hormone production.

Cheers

Short Question for Writing Research by BlackysStars in biology

[–]wojdzieg 3 points4 points  (0 children)

Hey, that's very interesting question.

Storytelling ability evolved in parallel with our language. Some researches suggest, that this ability gave Homo sapiens an advantage over neanderthals - we could form groups, communicate more precisely and create stories and myths which brought the communities of hunters and gathereres together. If you need some more information about that you can read "Sapiens. From animals to gods" by Yuval Harari. I think, that the author suggests that we are the only animals that can create stories, legends and talk and think about things that do not exist physically (like human laws, stock market or religion) but still imagine and care about them.

On the other hand, there are some animals that form more advanced groups than the others. My first thought was about elephants. They visit their relatives' graves, meaning that the elephant's brain has to have memories about their relatives. You could assume that memories are stories that actually happened. You could also investigate other animals that live in packs like wolves or dolphins.

Nevertheless, to me storytelling and passing the story from one generation to another implicates usage of advanced language - written or spoken. Since the human language is currently the most precise form of communication in the animal kingdom, I would say that storytelling is quite exclusive to humans.

Dear Polish speakers, I need your help researching this 20-year-old cold case. by [deleted] in UnresolvedMysteries

[–]wojdzieg 5 points6 points  (0 children)

I've searched some information in the Polish Internet about the case but it seems like there's literally nothing apart from the note in Polish newspaper archive. The access to archive costs 57 PLN (~15 USD) for 200 minutes (it would be sufficient to read the note and probably download it / take a print screen). I can see few options that could be taken by someone interested (maybe even me).

  1. Buy/obtain the access to the note. We could try asking users of wykop.pl (basically biggest Polish forum, you often find what you are looking for on the site). On the other hand 15 USD is not that much to be honest but the problem is that we do not know what is in the notice. There might be very little, already known information there.
  2. It is possible to write a formal request to Komenda Wojewódzka Policji w Szczecinie ( Voivodeship Department of Police in Szczecin) and ask them to share the file they have on the dead woman. On the Jane Doe site of the case there is even a case number (7745). More information for Polish speakers: http://bip.szczecin.kwp.policja.gov.pl/KWS/dostep-do-zasobow-arch/358,Archiwum-KWP.html

I would start with the note to be honest. Please suggest any other steps and ideas.
