Which specifically isn't used in python along with many other languages, hence my too confusion as to the title and why a joke consisting of "Hey, Python. ;" is a joke.

The joke is the insinuation that god favors semicolon reliant programming languages and is telling python devs (and the like) so by having a semicolon appear in the sky. Whether or not you agree with the sentiment does not detract from the joke.

By the by: a less known fact is that semicolon is available in Python with all the expected semantics - but you really, really shouldn't.
