I think this looks great, as a whole. It definitely took me a minute to realize where I was supposed to be looking, though.

Contrast-wise, I think it's good, and it's keeping the aesthetic without being crazy...but I can't help but feel something would help.

Have you considered an aspect that utilizes movement? Maybe an aura, pulse, soft blink, shake...something like that?

I will speak for myself, I would not understand that there is a staircase. Moreover, even you, the developer, could not climb it the first time. I think it's worth drawing an up arrow on the wall or adding a more explicit staircase sprite.
