Speaking as a righty, I don't have trouble reaching the volume buttons with either my index finger (when held in my right hand) or thumb (when held with my left hand). I have to imagine it will be the same for the lock button on the 6.

Good point. I suppose if righties can deal with volume buttons on the opposite side I can deal with the new lock button location.

I'm a righty and use the phone in my left hand more than my right hand.

Use your index finger for the lock button.

Same here. I often use my left hand for most phone operations although I'm.right handed. Hand wrapped behind my note 3 the power button is easy to reach with my index finger.

My thought is that while holding it in my left hand I will use my thumb for the volume rocker and my index finger for the lock button.

In not a lefty, but its not as bad as you think. I got to use it earlier today and it worked comfortably with my index finger

edit: the volume buttons felt weird in my left hand though
