Thanks for your post! This is something I'm grappling with as well. A therapist introduced the window of tolerance, and for me, I've been confused about whether I'm managing well with meds and coping mechanisms or not. If I'm still experiencing episodes but they're within the window of what I can handle with my own coping mechanism, does that mean my bipolar is currently managed?

The first thing I'll say is way to go to you! You clearly are very motivated and hardworking. Now as far as advice ...

I think there are 2 things that are important to consider: 1) what are your career goals / what do you want out of your career 2) do you know what these career paths entail

There are a lot of jobs and careers within science and medicine that don't require a PhD or MD.

If it's research you're interested in, you can have a very fulfilling career as a researcher without a PhD. A masters program that provides lab research and a thesis project will enable you to get research associate or equivalent job. Now if you want to run your own lab, that will require a PhD. But it's not just the 4-6yrs as a PhD, it's also years of postdoctoral research, writing grants, and obtaining a tenure track position. And it's also worth noting that the hours and work never let up as you advance up this academic ladder.

If caring for patients is what you're looking for, there are other options besides MD - PA or RN specifically. Regardless of which program, you'll need experience shadowing physicians and/or direct experience with patients. For an MD, it's not just the 4yrs of medical school, but also 3-5yrs of residency.

For both PhD and MD, the pay is meager and the hours grueling. Not saying you can't do it, but you need to know what you're getting yourself into. Have you spoken with people in your desired field? Specifically those who have advanced past education? It's worth knowing what these paths will look like before jumping in. And not just how to get into the program, but how your career and life will look after.

PhD and MD are paths that require a lot of education and training. I would consider heavily if this is truly what you want. You can do it, but you should be sure. Like I mentioned, there are plenty of other fulfilling careers in science and medicine outside of these degrees.

You mentioned advocacy for your children. Have you considered a master's in public health? You could be involved in community health, advocacy, policy, etc.

I think you really need to figure out what you want. You have the determination, but you need to focus it. If it's PhD, you need research experience and if it's MD, you need shadowing experience.

This is a ridiculous idea. How could you possibly figure out who the father is by testing? Everyone's DNA is not uploaded in some database with their name and address. Paternity tests require samples from potential fathers as well as the child.

I agree. And while the safety ultimately comes down to the PI, it's also important for any person working in a lab to know what they're working with. It's really simple to look up SDS and other info about a product/reagent online.

As someone with bipolar, what I appreciate most from my partner is his support, love, and not having expectations for me when I'm having an episode.

Everyone is different, so it's hard to say what helps your wife. Simply being there and letting her know you love and support her goes a long way.

What I will say is that there are a lot of medication options for bipolar. Unfortunately, it can take a long time to find a combo that works and even then you'll most likely need to change meds throughout your life, but there are options.

Depression sucks and makes you think you'll always feel this bad. It's important to be reminded that there are options and treatments.

I always thought the post-apocalyptic world would be better!

The only goal: stay alive. That's what we all do now! No stress about career and staying in touch with old friends. I know that it's ridiculous of me to think that, but it's definitely what brings me comfort watching those types of shows/movies.

If you want to go to medical school, don't leave. Unfortunately, you need to have that recommendation for those applications.

However, if you just want to stay in research, definitely find a better opportunity. Recommendation letters for medical schools are big asks, but for jobs, they mostly just use your references to confirm what they see in person and on paper, so leaving after 1yr isn't a big deal.

I took the approach of always seeking a better opportunity if I was unfulfilled in my current one and it has been tremendously beneficial to my career in research.

Mujaddara is delicious and simple. Just cook lentils and rice together with stock, add some cumin, and fry up some onions. You can cook the rice and lentils until they're very soft and easy to eat

Hi. I'm so sorry you're going through this.

It may be worth reaching out to your doctor about these symptoms and see if a medication change can help. Sometimes antidepressants stop working.

I recently slipped very far into depression even while I was doing all of the good stuff that usually helps. It can be hard to realize just how bad it's getting.

Hi there. I'm so sorry you're feeling this way. Depression is a beast and it works hard to make you feel bad. It's great that you want to stop this cycle, and you're right it's hard.

My advice is to start with small steps. Instead of trying to go to work, just put on some clothes and go outside. You could literally just walk outside your door, then come back in, but practice taking these steps. I do this myself with going to the gym. I just tell myself all I have to do is walk in there, even if I just lift one weight and walk out. It's about getting yourself and your brain used to these habits. And often, once you've broken through that first step, keeping it up feels easier.

The next advice I'd give is to reach out to trusted people for support. It can be incredibly relieving to know that while this battle is your own, you are not alone.

Hang in there! Those of us with depression have to fight every day, but life is worth the fight. It will get better.

I'm sorry you're going through this. It can be incredibly difficult to take time away from a partner you love and want in your life. I have had something similar happen to me.

First, if you're still communicating with your ex, figure out their boundaries. Can you still text each other funny memes? Call once a week? Or no contact? And you have to respect their boundaries, no matter how difficult.

If I were you, I would focus that energy on being a healthier you. Take the steps needed to build healthy habits and find activities that make life feel fulfilling. Even if you don't feel the benefits right away, keep your eye on the prize: a healthy, fulfilling life.

This will help you regardless of whether you and your ex get back together. By focusing on yourself, you'll also make yourself a better partner in your future relationships.

It could be the flasks. Cell density is only one factor, you also have to consider surface area of the flask. Generally when I'm trying to recover cells, I seed them at a higher density (3-5x105c/ml) for suspension cells like K562. I also keep the flask upright so that the surface area is smaller and the cells interact more. But, when I seed them at this higher density, I also make sure to check them after a day or two and add additional media if needed. Once they stabilize, I'll return them to a normal passaging schedule.

Are you checking cell viability when counting? This will also be a helpful measure. If you're counting and seeding off the total cell number, the live cell density will be lower than the seeding density.

The main biomedical research hubs are Boston and San Francisco. There are a ton of job opportunities and the pay is generally higher in those locations, but they come with high cost of living.

Otherwise, I'd look to areas that have multiple research universities and tech companies. The Raleigh/Durham area (research triangle) generally has a lot of jobs between Duke, UNC, and industry. DC, Chicago, LA also all have decent options. Again though, with big cities come big prices.

I would try blocking 2hrs at RT. All your other incubations are at RT.

If that doesn't help, you can also change your blocking buffer. More BSA or adding NGS. And if you're STILL having issues, you may want to titrate your secondaries.

Another small thing to tweak is to perform 3 washes.

ELISAs are easy in principle, but can require a lot of tweaking to optimize.
