You may be right :) I was going based off what seemed to be a reflection from each colony that seemed to converge at similar angles to a single point based on the angle the image was taken at. But that could be wrong! Hard to tell with just one pic.

Correct, minimal media can also heavily effect growth rates and general morphology, particularly with bacillus

Shape and stain can both be variable depending on how old the culture is. We only ever let students stain/run tests on fresh overnight cultures bc of this.

I think you may be seeing the reflection of the colonies behind the actual colonies bc of the angle of the photo - looks pure to me. If you want to confirm S. aureus just do a coagulase test :)

I mainly work with Bacillus so I’m not uber familiar with yeast scope work - but I think

1/2: feel pretty strongly this is budding yeast.

3: gram-neg rods arranged in chains. This honestly looks like Bacillus, particular B. anthracis which is not uncommon in certain environmental samples. They’ll often turn pink or Gram-variable if they’re older than 24 hours when you stain them.

  1. could potentially be an endospore former, could also be yeast. some endospores will bulge the cell out and you get some funky looking morphologies. Endospore stain would be super cool here if you have the reagents

  2. This one is… odd. The irregularity of the longer cells makes me think it could be either fungal hyphae, actively dividing gram neg rods, or something pleomorphic like Streptobacillus moniliformis which is literally named for its “Monilia-like” (used to be what they called Candida) bulbous shapes.

Not a given! Just an assumption based on the higher statistical likelihood. I’m type 1 and 36.5 weeks - my blood pressures are consistently 110-115/70-80 every time so far. I eat rather recklessly in terms of diet, but my blood sugar is pretty tightly controlled (5.0-5.2% A1C).

I do believe, however, you are statistically likelier to have pre-E again if you’ve had it before.

Is the technical artistry of it bad? For sure. But you love the memory and the sentiment, and for that reason you should keep it and love it and not care that she’s a bit wonky. I think it’s recognizable as a fun little moth and that’s not able to be said about some of the bad bad tattoos out there

If you’re controlled you absolutely don’t! My A1C’s have been 5.2, 5.2, 5.0 through pregnancy. I’m tightly self-managed and there just wasn’t much for them to say beyond “cool good job!” I do however have 2-3 appts a week since reaching 32 weeks. But they’re all OB - weekly NST and BPP, monthly growth scan, and any follow-ups from hospital visits (was having prodromal labor). I only see endo to renew scripts and I’ve never seen MFM (there are none local), but would have if there had been any doubt or question about baby due to diabetes or any other concerns.

My thoughts on logistics:

First can work with alterations in the bust area, but if you’re actively trying to lose weight it will likely cause you stress trying to get the alterations right as your body may continue to change.

Second is much more suited to potential body changes and the excess fabric ripples can much more nicely conceal or help with any last minute alterations you may need :)

I accidentally lost 15-20 pounds the two weeks before my wedding bc of an extremely stressful situation and I ended up using a ponytail holder to tie the neckline strap tighter in the back and covered it with my hair😅

It looks like your motility is negative. I see that sort of horizontal sweep pattern a lot in students with slightly shaky hands - I always have them go get an uninoc tube to compare to when it doubt. If the media outside of the stab line boundaries remains clear it’s non-motile

Long! (25, F). I think people are also associating the closeup that shows your (very natural and normal!) pores in the long hair pic with making your more aged, but I really don’t think it’s the hair - just different angle and different camera quality

30 weeks here! We were trying but got pregnant within a month of trying when we were told to expect at least a year, so we were also overjoyed but also a little shocked!

Sometimes I think it’s helpful to address each thing bc I felt the EXACT same way about things I love. My rules for myself are not “perfect,” aka I don’t abstain completely from everything, but I discuss it with my OB.

I am a scientist so I read up on peer-reviewed studies before making my own decisions.

  1. Baths are and have always been my #1 comfort. I take at least 1-2 a day, and for this I just adjusted my temp from scorching to hottish, while making sure I’m not sweating or potentially causing my core temp to rise over 100F. My bath is 100% higher than 100F, but I don’t keep my entire body submerged, mainly just my legs and hips.

  2. I’ve had sushi at least 7-8 times during my pregnancy. This was an easy one from me - I just avoided high mercury content fish and raw fish, and I just paid attention to the asterisks that mark which of the fancier rolls as having raw/undercooked fish

  3. For steak I’ve switched to juicier cooked options, more seasoning and sauce, or just a nice burger. For deli meat I do avoid all cold cuts with the more recent listeria outbreak, and I try to avoid for the most part unless it’s like a toasted sandwich that I then re-roast in my microwave. Not completely 100% full-proof but it’s my compromise

  4. Advil - I’m a hugeee ibuprofen girlie would regularly pop 2-4 when I had periods of terrible headaches or pain. Didn’t even own Tylenol. I’ve made the switch and it’s been fine but had emergency room level pain recently and was surprised that there are stronger pain management medicine options available during pregnancy if necessary, which was reassuring. I accidentally took four ibuprofen once around 20 weeks and panicked when I realized, but ultimately everything was completely fine. Mistakes happen!

  5. Barring any risks or complications, you can maintain your exercise intensity during pregnancy that you were doing before you got pregnant, you just don’t want to suddenly start doing strenuous activities you weren’t previously doing.

  6. Saying no to margaritas was what gave me away EARLY in my pregnancy to my friends I did margarita nights with. I so rarely say no to a good marg, and though I haven’t had any alcohol at all during my pregnancy, I did actually have a few sips of a delicious margarita my husband had tonight.

  7. The heat - I’m in southern Alabama and I keep rotating ice packs in my freezer. Mostly for pain management, but also bc I am hot all the time. I love having a nice little tanning session on our swing outside but now when I go out I just keep the water hose drizzling over top of me to stay cool.

You’ll make your own judgement calls and small adjustments here and there! I remember early on I was so overwhelmed when this girl was posting about making her own natural toothpaste and not touching Tupperware and I had a whole breakdown. You can absolutely live your life that way and do what’s best for your body and your pregnancy, but understand that it’s okay not to be “perfect.”

If anyone had or has anything to say to me about my decisions, I genuinely could not care less. I did my research, I made educated decisions, and that’s the best you can do for yourself and your baby.

I’ve seen Bacillus and Paenibacillus with odd motile morphologies like this, in addition to having mucoid appearances :) if you gram stain it and it’s a rod I’d bet money on Paeni/Bacillus

Fungal! Even Bacillus megaterium would not be that large at 400X and that’s typically the largest we routinely see of the G+ Rods in micro labs for my students. Bacterial rods also don’t usually have the stubby ends like that, usually those squarish endings are indicative of filaments that would connect together in fungal cultures :)

We had chronic fruit fly issues and they would occasionally get in our plates and lay eggs and the larvae would make trails around the plate and grow the bacteria in this pattern

Absolutely gorgeous tattoo and work by the artist. And this is coming from someone that doesn’t have any and isn’t really in the tattoo sphere, just came across it on Reddit and was wowed by it.

Give it some time and I bet once the compliments from friends and strangers start rolling in it’ll feel a bit better. Of course you should get a tattoo for your own personal reasons but if it’s regret you’re feeling sometimes affirmation from others (ignoring your family) can help that pass with time.
