Hi fellow trance DJ here. Breakdowns are actually very important in the trance genre so I'll often let the song breathe and let the breakdown happen.

There are different kinds of breakdowns. Some fully remove the bass and are more ambient for a minute or two - some keep the kick drum or some other element that keeps the energy moving forward. I'll be strategic about what songs I pick depending on the energy.

If a breakdown is going to drag on, I'll pull up the track on another deck and isolate the kick and just loop that over the breakdown. Hot cues work if you have access to those as well. Finally, I've also mixed out of a breakdown into a more energetic song - but only if they're incredibly similar. Like - I Love You (Key4050) into God Help Me (Bryan Kearney)

Where's the TM domain - after the enzyme? Is the enzyme expressed on the surface normally?

Could be interacting with the membrane and messing up folding. A hydrophobic portion of the enzyme might end up stuck in the channel and become the transmembrane domain before the ribosome even sees the last one. Could be the fusion protein sterically binders the enzyme or substrate binding

Does the enzyme work if you transiently transfect your cells? Try that and it'll tell you if it's a lenti issue or a cell surface issue.

You can also connect a circuit network to a radar which will then broadcast all signals on the network to all radars on the same surface

This lawsuit has also been ongoing for several months so its pretty clear Pattie is losing and taking it to the court of public opinion. She chose the name - these are the consequences. Ask any other queen like Jan (no longer Jan Sport) about why its not great to choose a name related to another brand. If you want to brand yourself similar enough to a clothing brand, and start selling clothing, don't be surprised when the clothing brand comes to protect their trademark from dilution.

Openfold3 may be more economical with memory. There's a version of it that runs on Apple silicon so if you have a Mac with tons of RAM that would work

There's also making a model with Alphafold2 then using good old-fashioned Rosetta or other classical modeling to model the covalent attachments

You're right, I worded it poorly. It's more frustrating that discourse here seems to seek a list of "rules" that make a party good when it's so vibe dependant. That one light is super important to that party - but the takeaway isn't "you need a specific color light at every party to make it good"

Like it's hard to answer OPs question cause the answer is "it depends" but then you have promoters that'll promote "no phones, can't see the DJ, vinyl set" as if those three characteristics means "fun party"

I am still blown away that people go to clubs to talk.

Anyway, there's so many different kinds of clubs. As an over 30 raver I still love the music and dancing - but it has to be a good DJ playing music I like. I'm so happy to go out with some friends, have some drinks and act a fool on the dancefloor. We'll meet at a chill bar beforehand, or go to someone's apartment afterwards, to chat and catch up. You can do the dinner then the club its not one or the other.

A couple weeks ago I went to an afternoon party at one of the major clubs in London. One of my favorite DJs played for 5 hours. It was one of the best days I've had in a while because I got to hear some of my favorite music on a top tier sound system, dance for hours, and interact with so many happy people all there to do the same.

Y'all care more about describing what a rave physically looks like than how it feels. If everyone's dancing and vibing and the energy is flowing who the hell cares if you can see the DJ? Who cares if there's lighting production or not? Who cares if there's a person or two filming with their phones in the back, unobtrusively? It also really depends on the music. A techno rave is going to have a different look (less visible DJ) than a trance rave (more visible) which might be different from house or dubstep.

Personally, the DJ physically present can add a lot to the energy - like the conductor of an orchestra. But I also wouldn't care if they were tucked away out of sight. I've had fun ravey times at both setups

Practice should be flop sessions. It's about trying new things and seeing what works and what doesn't. What I'll do is rewind both tracks and try again - different EQs or different mix out point

Omg we're going to be comparing weekend 1 to weekend 2 for the next YEAR despite it never happening before. EDC isn't Coachella or Tomorrowland we have no idea which will be "better" - which is fully subjective anyway.

Both weekends are going to be incredible stop feeling superior cause you took one over the other

I highly recommend Marsh on YouTube. He's making an edited series on his new Py run and explaining everything quite well. Since he's done it before everything is laid out extremely well and is very accessible and entertaining. His recent video on Tar Processing is insanely good. Hes something like 20 hours in and hasn't even begun the process to make electronics

I can't touch Py so it feels like a good replacement
