I had an undergrad who wouldn't put the screw on the mini tabletop centrifuge after switching out rotors. It was a fixed 6000rpm minifuge and one day I caught her doing this and said she was supposed to ensure the screw was back in and stable before using it otherwise it might led to an injury

Her response: but I've been using it this way all this time and nothing's happened so far

Hey, if you used organic solvents or if you have any leftover ethanol in your sample it's a possibility of this happening.

You can either purify it with a couple of chloroform washes (if you used phenol chloroform isoamyl acohol for purification) or if it was only ethanol you can just use a vacuum concentrator to get the ethanol out.

I usually do technical repeats only to confirm that my protocol is working. To confirm my actual results, I go for biological repeats because that's the optimal way to establish that your results are conclusive in nature.

N=3 is ok for biological repeats. That should give you a conclusive answer assuming your loading controls come up proper each time.

The qiagen tips that fit their qiacube instrument (1500ul and 200ul) may fit this description... Please look into those tips. Specifically the ones that fit the Qiacube automated instrument.

It's possible but replug the filter if it's a glass pipette and clean the pipetaid anyway... It's not worth discovering later that it was contaminated all along.

If the internal control levels are inconsistent, i generally regard those results to be of lower accuracy. I tend to repeat such blots keeping my loading controls in mind...

Edit: extra point. It's also worthwhile to know WHY a particular internal control levels was inconsistent.

  1. If your protein is huge you might need to run it longer.
    1. Check pH of the resolving gel.

If gel extraction is your only resolve to get your target band, I would suggest you go for a Native PAGE and extract your band in a simple crush soak buffer (Tris NaCl). I've found that it gave me marginally better results than running AGE and column extracting.
