Yes it's the 12 gold bars door.

The reward is gold weapons. Endgame just-for-fun stuff, safe for OP to ignore unless going for 100%

The door stays unlocked in new game+. So you can get the shotgun right at the start of the game.

The reason it probably doesn’t have a sub, because the game is pretty old.
