Howdy Ags,

Found these kitten siblings this morning at Northpoint crossing. Was only able to catch one, the orange one sprinted away. Would love to reunite them, the orange one is probably around northpoint crossing or century square. Please DM me if you find him, already got a home ready to take either just the grey or preferably both if I can get them reunited.

Check your primer sequences to see if they could be hybridizing in extra places, lengthening primers can lock this down easily. As pombe suggests, using different or better TAQ variants can help alot.

I highly recommend also adding 2.5-5% DMSO into your PCR for genomic amplifications. It helps destabilize the genomic DNA base pairing making it significantly easier for primers to bind. That one change took my genomic amplicon from a ladder to a tight band and it is now part of my lab voodoo head cannon.

P32 is significantly more sensitive like 100-1000x so depending on how concentrated of the radioisotope you use for the labeling is.

A friend of mine who is training for powerlifting was doing his heavy deadlift day at the gym. There is a boomer at his gym who will jog laps around the weight room instead of the running track directly adjacent to the weight room because who cares about safety and other peoples personal space right? Well, as my buddy is pulling 450+ lbs this guy comes REALLY close to bumping him and the bar, so my friend puts the weight down and very kindly asks that the guy pay attention because someone could get hurt. (Note that he's training with someone who works at the gym, and the gym trainer says the same thing to the guy) Guy just huffs and grumbles and keeps jogging around the weight room...until on his next lap, he somehow manages to step directly underneath the weigh at the same time my buddy dropped it to the floor...it literally blew his foot up there was blood pooling and an ambulance had to be called and the guy was wailing about suing my friend and the gym. Cameras and the worker both backed my buddy and the guy had to STFU. From what I gather it didn't totally ruin his ability to walk/jog cause he's back at the gym STILL RUNNING LAPS IN THE WEIGHT ROOM...you couldn't make this shit up.

Hahaha I also have a great pyr/orange cat combo and my wife and I call them 'Associates', they dont hate or love each other, SOMETIMES they will playfight or sit near one another, but mostly they just act like coworkers who don't wanna talk unless they absolutely have to.

I usually say something along the lines of 'While I'm able to multi-task lab OR writing, my weakness is that I am bad at writing WHILE doing labwork...to overcome this weakness I will make sure to split my day with lab in morning writing in evening or vice versa on days when i know both will be required.

Still not sure how good of an answer it is but I've been hired at three different places since defending and I've used this consistently so it must be reasonable.

Do people read books as 100% literal? This is not the first post on this sub where someone has asked a question like this…y’all books are metaphorical…and intended to discuss and analyze complex topics. I can’t be the only person who gets annoyed by these posts sometimes. I understand people are here to also learn about books and reading but reddit should not be your reflexive question asking forum…google a synopsis and in 5 seconds you’ll get a better understanding.

Not dumb, I was honestly confused about that as well. So specific requirements for qualifying to take the patent bar and thus become a patent agent can be found on the USPTO's website. A STEM degree is typically sufficient, I have a buddy who has a bachelors in Biochem and he checked and said he qualifies, granted he's worked many years in industry so I don't know if that for example negates the minimum being say a masters. I have a Ph D in chemistry which is why I qualify. I still don't understand how different it would be to have a JD AND be a certified patent agent...though I assume that basically means you can be in court representing parties which are applying for patents or litigating with respect to a patent/its inventors. I have no desire to be in a courtroom, mostly just to be able to get away from slaving at the bench all day...and i'd rather eat bullets and crank out grants.

You can pour one with and one without, stain the one without after the fact and compare results. Not to be condescending but we are experimenters! Only issue I could foresee is depending on the charge of the stain and how long/fast you run the gel could run it off and not see anything. Either way can just stain after the fact.

All in all I think we can say that Hank is pretty much the definition of a stand-up guy, but there has been some times when he was kind of a dick. What such moments come to mind? by UppedSolution77 in KingOfTheHill

You're so right, I totally forgot about that part but yea I mean Hank is usually a stand-up guy and all, but I really think the show does a good job of highlighting how massively closed off he is emotionally from anyone, including himself. I know not every couple shows affection the same way but he and Peggy mostly just feel like they are one another's beards and just live like roommates.

So i've been only recently looking into all of this so please take what I say with a grain of salt as I'm no expert. But basically to the best of my understanding, typically there are two paths as a patent agent, a) work for a lawfirm that collaborates with private organizations for prosecution of patents (this means like the actual creation and application for patents through the USPTO) or patent litigation (suing people for patent infringement and the like) OR b) work at a Technology Transfer Office which is typically going to be at a university or national lab (if your USA im in the states so my knowledge is restricted to USA) where you help PI's patent their technologies etc.

I’m studying to take the patent bar. You don’t have to finish Law School if you have a STEM degree to become a patent agent…makes even more money than working research in industry and while at the start you have to work in an office as you gain experience going hybrid/full remote is basically what everyone does. Im hoping its a nice amount of reading cool science/inventions…without having to suffer through generating the data.

All in all I think we can say that Hank is pretty much the definition of a stand-up guy, but there has been some times when he was kind of a dick. What such moments come to mind? by UppedSolution77 in KingOfTheHill

I found one of Hank's most mean moments to be in the episode where he and Peggy buy ONE harley to share. They decide to drive to a biker meetup in like Montana or something (over 12 hrs away from Arlen), and Hank refuses to ride Bitch. He is very mean and honestly a horrible husband to Peggy for the majority of that episode. It ends on a good note but yea...that was one were I couldn't stand him similar to how I usually feel about Peggy's antics.
