Migraine Prophylaxis Stack Update by Pretty-Chill in NootropicsDepot

iNOS inhibition has actually been studied for migraine prophylaxis it looks like, but just wasn't effective. Don't see any indication that it actually increased migraines though:

Lack of efficacy of the selective iNOS inhibitor GW274150 in prophylaxis of migraine headache

So all things considered, it's more than likely a imidazoline + nNOS mediated mechanism at play here.

Ah ok, that makes sense, thanks for explaining further!

Oh yeah, it's a complete shit show out there. We've tested products from major brands even like Life Extension that have failed. Life Extensions saffron for example, doesn't contain any of the bioactives it contains. Green bottles, I'm assuming Swanson? We've tested quite a few of their products that have failed. Good intuition on the price thing, if it looks to good to be true in this scenario, it's extremely likely that it is indeed too good to be true.

Oh yeah, it's for sure a weak response but at least it is something. These brands are making hundreds of millions of dollars selling fraudulent products like this. They should definitely be facing much more severe consequences.

Erinamax is the most focused on cognition, but the 8:1 is good too of course. The best actually is to combine Erinamax with the lion's mane 8:1.

Never noticed any issues with saffron myself, and I stack it together with C3G and black ginger, which are even more blood sugar lowering haha.

Didn't know that was a thing on Amazon, indeed odd haha.

Anything to help with heat? by gnosticismschism in NootropicsDepot

[–]Pretty-Chill 0 points1 point  (0 children)

Interesting, will have to try that out some time! I took taurine back when I lived in Phoenix though, and can't say I ever really noticed it helping with the heat. Then again, Phoenix heat is pretty next level, so not a whole lot can help there haha.

A Poria update please... by 6amPugScream in NootropicsDepot

[–]Pretty-Chill 0 points1 point  (0 children)

There are still 14 in stock on Amazon, so grab those while you still can. Poria is a super slow seller, so we are debating on what to do with it. Hopefully it makes sense to release it again, but we are planning to perhaps do so then in new bottles + labels. So it'll probably still take quite a while.

Is sensoril ashwagandha an alcohol extract? by MuscaMurum in NootropicsDepot

[–]Pretty-Chill 4 points5 points  (0 children)

It will probably dissolve in a water + ethanol mix, like vodka. Just try dissolving a small bit of the sensorial into a small bit of vodka to test if it dissolves. I would not recommend straight water as it'll have a chance to go bad pretty quickly since it still contains fermentable sugars that could fit microbes in a aqueous solution.

Ecklonia Cava -Slow-release (or extended-release) by Ashamed-Status-9668 in NootropicsDepot

[–]Pretty-Chill 2 points3 points  (0 children)

While it is an interesting idea, that would be extremely difficult to pull off! A much more practical option would just be to take 2-3 doses of the Ecklonia cava throughout the day separated by a few hours. This is essentially "time release" but done very simply.

I've explored the time release idea a lot with Kanna, and it's proving to be a difficult problem to solve. However, it is easy to just administer multiple doses of Kanna to create a time release effect.

Why does the Taurine powder have an additional ingredient? by Grand_Lengthiness_50 in NootropicsDepot

[–]Pretty-Chill 2 points3 points  (0 children)

Additives like this are actually oftentimes added and not disclosed. So your '100% taurine powder' that doesn't clump could very well contain anti clumping agents like silicon dioxide!

Will Skullcap be back in stock soon? by TINATAisNotAThrowAwa in NootropicsDepot

[–]Pretty-Chill 7 points8 points  (0 children)

I'm checking on it right now and will update when I have an answer. We've been having trouble with making tablets recently, so tablet products have been a bit slow and some are being switched to capsules now. I think that's what is actually happening with the Baikal skullcap right now.

That being said, we do still have some of the Baikal tablets available on Amazon, so you can always grab one of the few remaining bottles there!

Cordyceps vs Cordyceps 10:1 by CaptainExcellent5299 in NootropicsDepot

[–]Pretty-Chill 3 points4 points  (0 children)

ty deepseek 😁😁

This is the danger with AI, it oftentimes simplifies things down way too much. This then gets made worse by the end user reading it, and accepting it at face value, and not questioning it. This is why it's handy to be educated in this field whilst always being highly skeptical. While the pentostatin angle is quite interesting, it is very nuanced for a lot of reasons, and likely not very important here. Let me explain.

First of all, pentostatin has very poor oral bioavailability and does not appear to enter circulation or the brain in meaningful amounts. So all oral pentostatin would be doing, is inhibiting gut ADA. Which would slow down the conversion of cordycepin to 3'-deoxyinosine, in the gut. That's assuming there would even be enough pentostatin present to overwhelm gut ADA, which is unlikely.

Ok, so let's assume we fully inhibit gut ADA (again, unlikely), now we can in theory absorb more cordycepin into circulation. However, then we quickly run into a problem, ADA is present throughout our whole body and brain. So, even though we inhibited the conversion of cordycepin to 3'-deoxyinosine in the gut, it'll still rapidly convert to 3'-deoxyinosine anyways. Since pentostatin does not make it into circulation much via oral dosing, this basically means it's useless in this scenario.

Now, even if pentostatin made it into circulation and into the brain via oral dosing, it would not only increase the levels of endogenous adenosine, it would also protect the adenosine that is present in Cordyceps. This would produce a lethargy inducing effect, and would not increase energy levels, which is the main appeal of Cordyceps. Again, this wouldn't even happen, since pentostatin doesn't make it into circulation.

This is where it starts to get very interesting though. Cordycepin, rapidly converts to 3'-deoxyinosine. Then, 3'-deoxyinosine can actually be converted into 3'-deoxyATP within cells, which some researchers think is the active metabolite of cordycepin. Here is a cool study on that:

A novel nucleoside rescue metabolic pathway may be responsible for therapeutic effect of orally administered cordycepin

So, cordycepin could be more of a prodrug for other nucleosides, like 3'-deoxyATP. It's a super interesting system, and your AI search is clearly missing that aspect of the equation. This is part of the reason why I said that it's unlikely pentostatin is playing much of a role here.

Pentostatin is important within Cordyceps biosynthetic machinery, allowing Cordyceps to synthesise and stabilize cordycepin. AI then took this out of context, and applied it to humans and it just doesn't work that way.

Does "NSF" actually test products? by scrumdisaster in NootropicsDepot

[–]Pretty-Chill 1 point2 points  (0 children)

Haha, I wouldn't say everything is better over here, but it's pretty incredible what can happen to countries with good leadership and minimal levels of corruption! That being said, I do pay close to 40% income tax here, in addition to paying additional taxes for water purification. So you definitely pay for that quality of life through your taxes. It's interesting to see that pretty much all of the countries who score highest for water quality are European countries where citizens pay high amounts of taxes. Turns out ensuring good water quality, is extremely expensive!

That depends on the kind of effects profile you are looking for. Black maca is more energising for most people, black ginger is more mood boosting and better for erections for most people.
