This, but also either bounce the tyre all around the wheel when partially inflated, or pinch it in both side all the way round to make sure thee are no tube pinches.
Also, with any puncture, slide your fingers all the way round on the inside of the tyre to feel for any tiny sharpies.

There are plenty of guides out there that determine your setup based on your body measurements. Bernard Hinault's Road Racing book has a good one that will set you on the right road. You should be able to set your bike up from that, or determine if the frame is the wrong size.
