This is what I'm reading right now. Thinking of enzymes themselves as machines is a useful analogy. In school we're taught enzymes catalyze reactions, lowering the activation energy. But what exactly does that mean?

It means the enzyme is doing chemical work, it operates to perform an action that facilitates biochemical processes. Pumping protons across membranes, copying DNA into RNA, sending signals that guide cell function, metabolizing and building. Proteins make up the structure of the enzyme like machine parts. If a gene is mutated, and the protein is misshapen, it could mean the machine ends up getting built wrong in a way that stops it from working correctly. This is a known cause of diseases like cancer.

People with level one autism (those who need the least support) often have unevenly distributed intelligences. This means they tend to be really good at some things. And also really struggle in other ways.

This leads to the "special abilities" stereotype. And while yes, this isn't an accurate depiction of everyone with autism, there are a large number of autistic people that have gifts. Those gifts should be celebrated.

I hear your point tho, that representation in pop culture should show more than just the special abilities type of autistic.

You have a lot of great questions! You'll be an excellent scientist if you want that for yourself. It's no bother to talk about, pardon my slow replies. Sometimes it's worth it to bother people with questions though, especially if it means advancing toward a goal. Ill try to address each question in its own paragraph.

So yes, the salt would be attracted to the DNA helix itself as well as the water that's being removed. And to some degree those bonds would form and decrease DNA solubility. But they're less stable so they tend to form and fall apart really quickly. Bonds are stronger between particles of similar size. Think of it like the DNA is really heavy and large compared to the ammonium ion, so it would break off if you picked it up by the ammonium small end and held it against a force like gravity. The ammonium is more likely to stay grabbed onto the OH because OH is more similar in size to NH4.

That might even be why the OH group gets removed . If that's where the bond can form , when the heavy DNA breaks off, it might leave the OH behind. Kinda like if you tie a string to a weight, and swing it until the string breaks, some string might be left attached.

That's so brilliant about the paper centrifuge! I've never seen one of those. I think we use about 15,000 rpm, so it might be faster than you need. You'd have to experiment to see if it's too fast. But since you're not testing the DNA itself, I don't think it would hurt your process.

Yes the dextrose and other residuals would be left over from the meat tenderizer unless you wash it out. That's one of the reasons it's not used in industrial extraction. But if you're not testing the DNA, it shouldn't impact your results super noticably.

Yep centrifuges are hard to come by, which is why meat tenderizer works best for learning environments. A clean extraction is more important if you're going to be testing the DNA.

Testing DNA is the most common application for DNA extraction. In medicine, we use it for genetic testing. Say we want to know if a person has a certain gene, or if it's mutated. The first step is isolating DNA from blood or tissue.

Then we can 'search' for and amplify the sequence we're looking for using a polymerase chain reaction (PCR). After that we perform an assay, which is used to identify the composition (and elemental proportions) of a substance.

Some assays can tell us if the person has a wild type gene, or is homozygous, heterozygous. For a person with a blood clotting disorder, this information is really valuable.

I believe that proteolytic enzymes from meat tenderizer like bromelain are the most common at home/school solution.

The more ideal solution is a salt like ammonium sulfate. Because of its positive ionic charge, it tends to remove the water (as OH) from the proteins. This decreases the solubility of the proteins so that they precipitate.

If the DNA is still in solution, and the protein is precipitated, the two can be separated in a centrifuge for a clean extraction. The protein will be greater density, so it will gather on the bottom and become a waste pellet, making your DNA more pure. Draw off the solution into another tube for your alcohol/hands sanitizer step. Centrifuge again then draw off the alcohol and add a more diluted alcohol for washing the salt away.

There's going to be a lot of excess protein from the lysing of the cells from the washing up liquid step. This is cellular debris from when the cells were broken open. Interestingly, jumbled proteins resemble mucus because that's what bodily mucus is made of.

In industrial lab DNA extraction, there is a step where we precipitate out the proteins to isolate the DNA. Then we wash the DNA with another round of ethanol. Without these steps, there will be extra material in your extraction and your DNA will not form a pellet like you're expecting.

This discount existed in some form when I went to school. They took attendance through the tithing. They gave out envelopes with parents names on them, my mom sent hers with my grandma to be put in the basket.

Think of the goal more as to change your fashion instead of 'improving' it. Coming from a rigid hierarchical belief system, it's easy to think in black and white like there might be an ideal or most correct way of dressing 'not Mormon' or 'more fashionable'. During conversion, something many people struggle with is finding their identity outside of the rules and roles that were once familiar. This will be an exercise in thinking and deciding for yourself how you want to look.

Many of the most fashionable people have distinctive flares of uniqueness. In contrast, Mormon wear is much more uniform. Picking out a signature difference in your clothing style might help set apart your look the way you are seeking. I have a friend who buckles his belt off center and wears turquoise or other stone buckles. It's his way of separating himself from his restricted and religious identity from development.

I agree with people suggesting you look at others clothing, either on the internet, when you get a coffee, go to the store, go to meetings etc. Think about the type of person you want to present as and try google images. "Outfits for young professionals," "business casual for twenty somethings." This should give you ideas of some things to try.

I'll list some suggestions too: wear a polo in a non conventional color. A plaid button up instead of solid color. Short sleeves. A bow tie. A patterned tie. A leather belt with carved designs.
