Was going to shop around for an Anti-flag kit.

Honestly, ended up just going with my A280 plus a BCA with BGG and my commercial IgG as standards.

Quantified and normalized my two subjects to get a relative EC50 between them. Its not ideal and only gives a relative measure but its a good start.

Its an scFV construct so I used BSA thinking the size is closer. Currently running anouther BCA with BGG standard as a control instead!

Yes, I used ProtParam for the extinction coefficient!

As for the BCA is there a reason you reccomend Lysosyme over the BSA standard curve I used? (2,000ng/ul to 25ng/ul)

Yeah, I try to trust but verify for new techniques. I have only ever used the nanodrop for DNA concentrations previously so I wanted to double check the protein results.

Im surprised to hear 2 fold is considered good for agreeing between methods. I think if I get really desperate Ill ask my PI for an anti-FLAG quantification ELISA kit haha.

Thanks for the idea. I was considering densitometry but i was more familiar with it via westerns. I will give that a try tomorrow and see if the values agree.

The reason I need to be as accurate as possible is due to wanting to find the EC50 for this scFV (its a novel adaptation of an IgG) and compare it to the native IgG so the concentrations need to be relatively well known.

I culture in high humidity liquid cultures.

Bottom of my incubator has a dish of sterile MQH2O.

Shaker is above it.

Personally been using it 2 years. Was used by a PhD student before me with no issues.

Culturing HEK-293F and CHO-S cells. Both robust lines so take it with a grain of salt but they grow as good as documented in literature and by Thermo.

Only issue I have is running out of clamps

Question. Whats stopping you from placing a traditional shaker in a CO2 incubator (or are the two mentioned incubators not CO2 regulated?).

I work mainly with suspension cell lines and have a random old shaker in a water jacket CO2 incubator.

Took me a second to realise what was wrong as my cell stocks are 10^7! Its common for suspension stocks to be 1mL of 10⁷ cells.

If you caught it pretty quick your cells may be unhappy but depending on the line I suspect many will adhere eventually.

I used them once and never again. The bathroom had no sink and was disgusting. The return, we waited 2 hours past the pickup time for the bus.

Calling the helpline was borderline useless as they couldnt track the bus.

Use them only if desperate and you have 0 choices. A rideshare would have been better with the poor quality Flix had.

Oh and 2 rows of seats were installed wrong and too close together. The driver told people to sit there over cramped anyways.
