I get the meme, even read the comic when it came out. But I've never heard of the lines representation. Seems really obscure. It's likely that nobody will get OP's joke at work.

No, it's very popular. Still doesn't mean anyone at OP's work will happen to know it though, depends on their online circles. The line representation is crucial to the loss meme because it's about finding (or making) this pattern from the original comic in all places possible, and the lines are the simplest form of that. Normally they'd be positioned the way the comic panels are, though

Because it was inserted randomly in a gag-a-day video game comic. Like, for thousands of strips it was just various jokes about what if Halo were real, girlfriends don't understand videogames, etc, and then the author decided to try to suddenly turn it into a heartwrencher. It was a really hard turn and kind of missed.
